FoF1-ATP synthase/ATPase in the parasitic protist, \kur{Trypanosoma brucei}

This thesis primarily focuses on the FoF1-ATP synthase/ATPase complex in the parasitic protist, Trypanosoma brucei. Instead of its normal aerobic function to synthesize ATP, it is required to hydrolyze ATP to maintain the m in the infective bloodstream stage of T. brucei and the related parasite, T. b. evansi. To better understand the composition, structure and function of this druggable target, my work focused on deciphering the function of three of the unique Euglenozoa specific subunits that comprise this complex molecular machine. Furthermore, the ADP/ATP carrier, which provides substrates for the FoF1-ATP synthase/ATPase, was functionally characterized and evaluated if it is physically associated with the complexes of the oxidative phosphorylation pathway. This thesis is composed of two topics, for which trypanosomatids and evolution are common denominators. First part deals with phylogenetic relationships among monoxenous trypanosomatids, with emphasis on flagellates parasitizing dipteran hosts, analyzed mainly from biogeographical and evolutionary perspectives. Second part focuses on the trypanosomatid Trypanosoma brucei, causative agent of severe diseases, which serves as a model organism for functional studies of evolutionary conserved mitochondrial proteins, in particular those involved in replication, maintenance and expression of the mitochondrial genome, also termed the kinetoplast. This thesis identified the mtHsp70/mtHsp40 chaperone machinery as an essential component of replication and maintenance of the kinetoplast, and also identified numerous conditions under which mtHsp70 has a tendency to aggregate. Moreover, several conserved proteins, previously identified to be part of the mitochondrial ribosome, were shown to be important for translation of the mitochondrial transcripts. Usage M. E. Levine model in clinical and community care in patients with colorectal cancer

The aim of the dissertation was to find out the specifics of nursing care in clinical and community practice with patients with colorectal cancer using the model of M. E. Levine. Furthermore, to find out which of these specifics are missing in the mentioned nursing model and to modify the nursing documentation according to the chosen conceptual model of M. E. Levine. In the empirical part we used a combination of qualitative and quantitative research which was carried out as part of the grant project GAJU 048/2015/S. The pre-research, conducted interviews with 8 informants was used to identify the key data for answering research questions and creating a custom-designed questionnaire. A non-standardized questionnaire was distributed across the Czech Republic to 300 patients with colorectal cancer. We also designed nursing documentation, which was verified by focus group with 35 nurses from clinical and community practice. The results show that patients are limited in areas of contact with friends, normal traffic, traveiling, and fear for themselves and their loved ones to maintain social integrity. In the principle of maintaining structural integrity, problems with diet, alcohol consumption, diarrhea, current treatment management, stoma and stoma type have been identified. In the field of energy conservation the influence of length of diagnosis and colonoscopy performed as part of preventive examination, pain sensation, and enough energy for normal daytime life have been confirmed. In the principle of conservation personality integrity, we have found that the length of time determines the fear of getting stoma, learning individuals with the same disease, experiencing negative feelings, and limiting private life. The work provides a comprehensive framework of the issue of patients with colorectal cancer. It provides evidence of the possibility of using M. E. Levine's conceptual model in colorectal cancer patients. Cílem disertační práce bylo zjistit specifika ošetřovatelské péče v klinické a komunitní praxi u pacientů s kolorektálním karcinomem s využitím modelu M. E. Levine. Dále zjistit, která z těchto specifik chybí ve zmíněném ošetřovatelském modelu a modifikovat ošetřovatelskou dokumentaci dle vybraného koncepčního modelu M. E. Levine. V empirické části jsme využili kombinace kvalitativního a kvantitativního výzkumného šetření, které probíhalo v rámci řešení grantového projektu GAJU 048/2015/S. Předvýzkum realizovaný rozhovory s 8 informanty posloužil ke zmapování stěžejních údajů pro zodpovězení výzkumných otázek a tvorbu dotazníku vlastní konstrukce. Nestandardizovaný dotazník byl distribuován v rámci celé České republiky 300 pacientům s kolorektálním karcinomem. Dále jsme navrhli ošetřovatelskou dokumentaci, která byla ověřována formou focus group s 35 sestrami z klinické a komunitní praxe. Z výsledků vyplývá, že pacienti jsou v rámci zachování sociální integrity omezováni v oblastech kontaktu s přáteli, v běžné dopravě, v cestování a mají strach o sebe sama a své blízké. V principu zachování strukturální integrity byly zjištěny problémy se stravováním, konzumací alkoholu, průjmem, zvládáním současné léčby, stomií a druhem stomie. V oblasti zachování energie byl potvrzen vliv délky obeznámení se s diagnózou a kolonoskopie prováděné v rámci preventivního vyšetření, pociťování bolesti a dostatku energie k běžnému prožití dne. V principu zachování osobnostní integrity jsme zjistili, že délka doby onemocnění ovlivňuje strach z vyvedení stomie, poznání jedinců se stejným onemocněním, prožívání negativních pocitů a omezení v soukromém životě. Práce poskytuje ucelený rámec problematiky pacientů s kolorektálním karcinomem. Podává důkazy o možnosti využití koncepčního modelu M. E. Levine u pacientů s kolorektálním karcinomem. Effect of submerged macrophytes on the structure of lake ecosystem and trophic relationships among fishes

This Ph.D. Thesis is focused on the effect of submerged vegetation on the structure of lake ecosystem and trophic relationships among fishes. Succession of submerged vegetation is a dynamic process and the vegetation community may change dramatically even during a year. Many environmental factors have impact on the succession of submerged vegetation and consequently the presence of the vegetation has substantial impact on the aquatic ecosystem. Nutrient level (mainly nitrogen and phosphorus) plays a key role in the succession of freshwater ecosystems. Further, herbivory significantly influences species composition and biomass of macrophytes and macroalgae. Herbivory of fish is a foraging strategy that is dependent on temperature. Foraging preferences of fish may suppress the palatable species. The rate of herbivory affects plant growth patterns and thus inpalatable species may be advantaged in the competition among plants. In aquatic ecosystems, herbivory is often performed by omnivorous species. The diet preferences of omnivorous fish differ among aquatic ecosystems and submerged vegetation can shape their trophic niches substantially. Biodiversity patterns of butterflies and moths on Mount Cameroon

Biodiversity patterns of butterflies and moths are investigated on the southwestern slope of Mount Cameroon, the only continuous elevational gradient of near-pristine forests in the Afrotropics. Three distinct seasons between 2014 and 2017 are sampled by two standardised methodologies (i.e., bait-trapping and manual catching of selected groups of moths at light). First, the effects of seasonality and forest habitat associations on butterflies and moths are investigated in the lowland rainforest. Then, the impacts of elephant-made natural disturbances on tree, butterfly, and moth diversities at mid-elevations is evaluated. The thesis also includes faunistic records of species never reported from the area, combining them with life history notes on selected taxa and species description of nine new species of Alucita. Finally, the last chapter focuses on the biodiversity of birds, trees, and butterflies in the last large patch of coastal forest of the studied area, and discusses its current conservation status. Life And Work of Konrád Maria Kubeš, T. J. (1890-1967): Missionary, Preacher, Excercitator and Deep Devotee of the Marian Cult

This dissertation in its first part inquires the life and work of Konrád Maria Kubeš (1890? 1967), a people?s missionary, a preacher, an excercitator and a deep devotee of the Marian cult. It is introduced with the socio-political history in 1890?1967, with the overview of the Popes and theological thinking between 1878 and 1978. Next, it looks in detail into each period of Kubeš?s life, his family background, high school studies in Hradec Králové, his theological studies in Prague, his entry into the monastic order and his activities in Society of Jesus. The dissertation pursues Kubeš?s life story in communist centralization monasteries, in Moravec, his short work in Bukovinka and finally his forced stay in Břežany near Znojmo. Afterwards, it analyses Kubeš?s theological work published in books and journals, as well his historical contribution. The final part of the dissertation evaluates Kubeš?s personality and his significance in the context of the Czech religious and monastic history of the 20th century. The author based his thesis primarily on archival recources, Kubeš?s short autobiographical memories, oral and written witnesses of his contemporaries and last but not least on secondary literature. The second part of the dissertation contains 182 text apendixes and 2 pictorial apendixes which are make up an indispensable and logical supplement to the whole picture of the life, work and contribution of Konrád Maria Kubeš. Disertační práce se v první části zabývá životem a dílem jezuity Konráda Maria Kubeše (1890?1967), misionáře, kazatele, exercitátora a hlubokého mariánského ctitele. Je uvedena společensko?politickými dějinami v letech 1890 až 1967, přehledem papežů a teologického myšlení v letech 1878 až 1978. Dále si podrobně všímá jednotlivých životních etap Kubešova života, jeho rodinného prostředí, středoškolských studií v Hradci Králové, teologických studií a kněžského působení v Praze, vstupu do řehole a jeho činnosti v řádu Tovaryšstva Ježíšova. Sleduje Kubešovy osudy v komunistických centralizačních klášterech, na Moravci, krátké působení v Bukovince a nakonec nucený pobyt v Břežanech u Znojma. Následně rozebírá Kubešovo teologické knižní a časopisecké dílo a jeho přínos tehdejší i pozdější době. V závěru disertace je všeobecné zhodnocení a význam Kubešovy osobnosti v rámci českých církevních a řádových dějin 20. století. Téma disertační práce je zpracováno především na základě archivních pramenů, Kubešových drobných autobiografických vzpomínek, ústních i písemných svědectví pamětníků a v neposlední řadě za použití sekundární literatury. Druhá část disertační práce obsahuje 182 textových příloh a 2 obrazové přílohy, které jsou nezbytným a logickým doplnění celkového obrazu života, působení a přínosu osobnosti Konráda Maria Kubeše. Parenting in the context of DLD - Parental experience in connection with difficulties of the child

The dissertation focuses on the psychosocial impact of developmental language disorder on a family and particularly parental life, namely the quality of parental life. In six chapters the theoretical part of the thesis presents the current view of specific language impaiment (developmental language disorder, DLD), its diagnostics and the therapy for it, as well as the system of support for children with DLD in the Czech Republic and the issue of parenting of a speech handicapped child. The goals of the empirical part were to give a description of the parental hardships caused by DLD in a child, identify the affected life dimensions, define ways of providing effective support for these parents and describe its impact on the parents' competencies, experience and ways of coping. Qualitative methods have been used in the empirical part of the thesis. The exploratory set for the study was made up of 25 parents (6 fathers and 19 mothers) from 20 families. A family's inclusion in the research was conditioned upon a diagnosis of DLD in at least one child of the family by a qualified specialist. The ages of the children ranged from 3 to 13 years, and school age children and preschoolers were represented roughly evenly. The parents were interviewed via semistructered in-depth interviews. They were given a questionnaire created by the author that was to show the characteristics of the participants and their families. Some of the participants also filled in a brief diary chart. The data obtained through the semistructured interviews have been processed though ATLAS.ti software and transformed into a grounded theory. The research has shown the complexity of the hardships experienced by parents and families when they have a child with DLD. The hardships experienced by these parents are in most aspects comparable to those experienced by parents of children with more serious handicaps, though their features dynamically change according to the child's age and the progress of his/her symptoms. During preschool age the main problems are the child's speech difficulties and various aspects of caring for the child. On the other hand the development of specific learning and behaviour disabilities and socialization disorders is the dominant problem when the child reaches school age. Further the research has shown the crucial role of providing support to parents of DLD children as a way to prevent failures in the parenting of such children. Four types of support provision have been identified based on different levels of expertise and the personal contact between the provider and the person receiving the support. Expertise, awareness, individual characteristics and the support provision system were identified as the intervening variables. The main characteristics of effective support provision were found to be orientation, the time-aspect, accessibility and communication. The consequences of support provision for parental coping suggest the importance of combining different support types and the significance of parent-provider dynamics. In the parental coping strategies five types of strategies have been identified in accordance with parental activity and the impact of the support. Disertační práce se zabývá psychosociálními vlivy vývojové dysfázie na život rodičů a potažmo rodiny dětí s vývojovou dysfázií, tj. na kvalitu jejich života. Teoretická část práce sestává ze šesti kapitol, ve kterých je představen současný pohled na poruchy vývoje řeči, jejich diagnostiku a terapii, podrobně je popsán systém péče o dítě s narušeným vývojem řeči (vývojovou dysfázií) v ČR a dále shrnuta problematika rodičovství v souvislosti s řečovým handicapem dítěte. Cílem empirické části bylo zmapovat zátěže, které vznikají v životě rodičů dětí s vývojovou dysfázií v důsledku obtíží dítěte, identifikovat oblasti života, ve kterých se projevují, charakterizovat efektivní poskytování podpory rodičům těchto dětí a popsat jejich důsledky na rodičovské kompetence, prožívání a zvládání. V praktické části byla využita kvalitativní metodologie. Výzkumný soubor tvořilo 25 rodičů (6 otců a 19 matek) ze 20 rodin, podmínkou zařazení do výzkumu byla odborně potvrzená vývojová dysfázie (resp. narušený vývoj řeči v předvýzkumu) alespoň u jednoho dítěte v rodině. Věk dětí se pohyboval mezi 3 a 13 lety, školní a předškolní děti byly zastoupeny přibližně rovnoměrně. S rodiči byly vedeny polostrukturované hloubkové rozhovory. Součástí výzkumu byl dotazník vlastní konstrukce charakterizující respondenta a mapující rodinné prostředí, dále někteří rodiče zpracovávali heslovitý zážitkový deník. Data získaná v rozhovorech byla s pomocí analýzy v programu atlas.ti zpracována a přetvořena do podoby zakotvené teorie. Výzkum ukázal na komplexnost zátěží vznikajících v důsledku vývojové dysfázie dítěte v životě rodiče a rodiny. Zátěž rodičů je ve většině aspektů srovnatelná se zátěží vznikající v důsledku těžších handicapů, její charakter se však dynamicky mění s věkem a vývojem stavu dítěte. V předškolním věku převažují zátěže související s řešením řečových obtíží dítěte a aspekty každodenní péče o dítě. Ve školním věku pak dominuje otázka školní úspěšnosti, socializace dítěte a rozvoje specifických poruch učení a chování. Dále výzkum ukázal na zásadní roli poskytování podpory těmto rodičům jako prevence selhávání péče o dítě s vývojovou dysfázií. Byly identifikovány 4 typy poskytování podpory podle odbornosti a osobního kontaktu poskytujícího s podporovaným. Jako intervenující proměnné se projevily odbornost a osvěta, individuální charakteristiky poskytujícího a systém, ve kterém k poskytování dochází. Jako hlavní charakteristiky poskytování efektivní podpory se ukázaly zaměření, časovost, dostupnost a komunikace. Důsledky poskytování podpory na rodičovské zvládání nasvědčují důležitosti kombinace různých typů využívané podpory a dynamiky kompetencí rodičů na jedné a poskytovatelů podpory na druhé straně. V rodičovských strategiích bylo pojmenováno 5 typů strategií podle aktivity rodiče a vlivu podpory. This thesis aimed to determine the influence of landscape conditions on the occurrence and movement of wildlife and to evaluate the success of game detection using different methods, focusing on roe deer. Roe deer are our native game and are now one of our most widespread game animals. The entire dissertation was conducted to propose the most optimal way of searching for a game on agricultural land in front of hayfields using unmanned UAV technology with a thermal imaging camera. In order to achieve the aim of the thesis, several sub-steps were carried out, which mainly dealt with the issue of comparing the methods of deer detection in forage crops (ground survey versus the use of UAV technology) and the issue of returning the deer to the crops after they had been flushed out or carried away. Other sub-analyses included evaluating the effectiveness of alternative methods of scaring deer from stands using different types of scarecrows and assessing the occurrence of red deer on the land concerning the location of landmarks. The solution of the dissertation thesis resulted in the proposal of a methodological guide for optimizing the search for deer on forage plots on arable land and permanent grassland. Tato práce si kladla za cíl stanovit vliv podmínek v krajině na výskyt a pohyb volně žijící zvěře a vyhodnotit úspěšnosti detekce výskytu zvěře různými pracovními metodami, se zvláštním zaměřením na srnčí zvěř. Srnčí zvěř je naší původní zvěří a dnes patří k naší nejrozšířenější zvěři spárkaté. Celé řešení disertační práce vedlo k navržení nejoptimálnějšího způsobu vyhledávání zvěře na zemědělských pozemcích před senosečí pomocí bezpilotní UAV technologie s termokamerou. Pro dosažení cíle práce bylo provedeno několik dílčích kroků, které se zabývaly především problematikou porovnání způsobů detekce srnčí zvěře v porostech pícnin (pozemní průzkum versus využití UAV technologie) a problematikou návratu zvěře zpět do porostů po jejich vyplašení, respektive vynesení. Dalšími dílčími analýzami bylo vyhodnocení účinnosti alternativních způsobů plašení zvěře z porostů různými typy plašičů a zhodnocení výskytu zvěře na pozemcích s ohledem na umístění tzv. orientačních bodů. Z řešení disertační práce vyplynul návrh metodického návodu pro optimalizaci vyhledávání srnčí zvěře na pozemcích pícnin na orné půdě a trvalých travních porostů.
