Cell cycle arrest as a hallmark of insect diapause: Changes in gene transcription during diapause induction in the drosophilid fly, Chymomyza costata
ŠIMŮNKOVÁ, Petra
2014 - anglický
The changes of relative mRNA levels of seven different genes, coding for key cell cycle regulatory factors (Cyclins D and E, kinases Wee1 and Myt1, Phosphatase Cdc25 (String), Dacapo (p27), and Pcna) were performed using qRT-PCR method. Two reference genes (Rp49 and ?-tubulin) served as a background. Significant transcriptional response to photoperiodic transfer were observed for two genes. While the relative levels of Dacapo mRNA increased during the rapid entry into G2 arrest, the Pcna expression was significantly downregulated during the beginning of G0/G1 arrest. Moderate transcriptional upregulations of the genes coding for two cell cycle inhibitory kinases, Wee1 and Myt1 accompanied the entry into diapause. The other genes were expressed equally in all photoperiodic conditions. Plné texty jsou dostupné v digitálním repozitáři JČU.

Dynamic karyotype evolution and unique sex determination systems in Leptidea wood white butterflies
VOLENÍKOVÁ, Anna
2015 - anglický
Chromosomal rearrangements such as fusions and fissions play an important role in promoting and maintaining speciation. To study these phenomena, genome architecture of three cryptic species of Leptidea wood white butterflies was investigated by means of standard and molecular cytogenetics. High variability in chromosome number and localization of cytogenetic markers was revealed, suggesting dynamic karyotype evolution in these species. Moreover, unique sex-determination system with 3-4 W chromosomes and 3-4 Z was found, possibly playing a role in the speciation process as well. Klíčová slova: Motýli; Lepidoptera; bělásci rodu Leptidea; karyotypová variabilita; fluorescenční in situ hybridizace; chromosomální přestavby; pohlavní chromosomy; speciace Plné texty jsou dostupné v digitálním repozitáři JČU.

Species pool size and realized species richness affect productivity differently: A modeling study
Current and expected decline in biodiversity have motivated a number of experiments studying how biodiversity affects ecosystem functioning. The positive relationship is usually found in experiments where species pool is manipulated; the relationship between productivity and realized species richness does not show any single trend. We constructed a simple competition model for a plant community based on the classical LotkaVolterra equations, with randomly generated parameters. We varied the species pool size and intensity of competition (range of competition coefficients). Then, we compared two measures of diversity used as predictors of productivity: (1) the size of the species pool and (2) the realized species richness, i.e. the number of species that remained in the system after competitive exclusion. Simulation results showed that productivity was always positively affected by the size of the species pool. With increasing species pool, both the selection effect and complementarity increase. The relationship between realized species richness and productivity was extremely weak within a set of simulations with a fixed species pool (i.e.where the diversity gradient was caused only by differences in the randomly generated parameters). The relationship between realized species richness and productivity was slightly positive for small species pool sizes and slightly negative for larger species pools. A species with high carrying capacity within the generated set of species usually decreases the chance of other species to survive but increases the productivity of the mixture, leading to negative diversity productivity relationship. On the contrary, presence of highly complementary species (i.e. species with low mutual competition coefficients) increases both, the realized species richness and productivity, leading to positive diversity productivity relationship. These two effects mostly counterbalance each other. These trends are not affected by the competition intensity.

Being angry, singing fast? Signalling of aggressive motivation by syllable rate in a songbird with slow song
JAŠKA, Pavel
2015 - anglický
In this study, we focused on the role of syllable rate in territorial interaction of chiffchaff males. We confronted each tested male with one of three playback types which differed in song rate (slow, non-manipulated, fast). The behaviour and the song of each subject were recorded before, during and after the treatment. Males that attacked the loudspeaker during playback sang faster before the stimulation. When birds continued singing during playback they increased syllable rate when exposed to fast or non-manipulated song. Our results indicate that syllable rate could be a signal of aggression in chiffchaff and play a role as a conventional signal rather than a handicap signal. Mice with different susceptibility to tick-borne encephalitis virus infection show selective neutralizing antibody response and inflammatory reaction in the central nervous system
PALUS, Martin
2015 - anglický
To address the underlying factors involved in development of tick-borne encephalitis (TBE) we developed an animal model of TBE based on BALB/c-c-STS/A (CcS/Dem) recombinant congenic mouse strains showing different severities of the infection in relation to the host genetic background. TBE virus growth, neutralizing antibody response, key cytokine and chemokine mRNA production and changes in mRNA levels of cell surface markers of immunocompetent cells in brain were measured in mice with different susceptibilities to TBE virus infection. Generated data indicate that the genetic control is an important factor influencing the clinical course of TBE. Klíčová slova: tick-borne encephalitis; recombinant congenic mouse strains; neuroinflammation Plné texty jsou dostupné v digitálním repozitáři JČU.

Physiological and biochemical responses to cold and drought in the rock-dwelling pulmonate snail, Chondrina avenacea.
ROZSYPAL, Jan
2013 - anglický
We assessed the capacity of rock-dwelling snail, Chondrina avenacea to tolerate environmental stress during seasonal alterations of activity and dormancy. We examined whether hibernation and aestivation share some physiological and biochemical traits, which can underlie potential development of cross-tolerance (i.e., high cold tolerance during estivation or high drought tolerance during hibernation). For this purpose, we recorded seasonal changes in the levels of cold, drought and anoxia tolerance and changes in basic physiological and biochemical parameters which are linked to dormancy and stress tolerance such as supercooling capacity, mass and hydration, rate of exchange of respiratory gases, total amount of proteins, lipids and glycogen, and metabolomic composition. Plné texty jsou dostupné v digitálním repozitáři JČU.
