Dynamics of digestive proteolytic system during blood feeding of the hard tick Ixodes ricinus

Dynamics of the main digestive peptidases involved in digestion of host blood in the hard tick Ixodes ricinus were studied. Gene expression profiles of the particular enzymes were determined by qRT-PCR. Enzyme activities were determined using specific fluorescent probes. Midgut morphology during blood feeding was observed and immunolocalization of the main abundant peptidase - Cathepsin B - was performed. The thesis consists of two studies contributing to knowledge about the ecology of two model saproxylic species dependent on tree hollows. It includes, firstly, an analysis of the habitat requirements of the violet click beetle, Limoniscus violaceus, and secondly, an analysis of the association of several saproxylic beetles with the presence of the hermit beetle, Osmoderma barnabita, in hollows. Chronic lymphocytic leukemia is the most common type of leukemia in adults in western countries. Despite the continual improvement of diagnostic methods and treatment, it still remains incurable. One of the main characteristic features of CLL is its clinical and biological variability. In present, the analysis of prognostic genetic markers is the most useful tool for understanding biology and clinical course of the disease and consequently the individual type of treatment. The presence of a NOTCH1 mutation is connected with shortened survival and resistance to conventional chemo-immunotherapy. In this thesis, we examined CLL patients with c.7541_7542delCT NOTCH1 mutation and the most sensitive and specific method was identified. We compared nowadays most commonly used methods: allele-specific PCR, fragment analysis and Sanger sequencing. The recent population declines of annual hemiparasitic Rhinanthus species may be a result of changes in mowing dates associated with the intensification of grassland management, but the causal mechanisms are not well understood. This thesis describes the dynamics of Rhinanthus spp. regeneration after cutting and of fruit ripening under silage or hay making, and suggest recommendations for conservation of the target species. Adipokinetic hormone activities in insect body infected by entomopathogenic nematode

The role of adipokinetic hormone (AKH) in the firebug Pyrrhocoris apterus adults infected by the entomopathogenic nematode (EPN) Steinernema carpocapsae was examined in this study. It was found that co-application of EPN and AKH enhanced firebug mortality about 2.5 times within 24 h (from 20 to 51% in EPN vs. EPN + AKH treatments), and resulted in metabolism intensification, as carbon dioxide production in firebugs increased about 2.1 and 1.6 times compared to control- and EPN-treated insects, respectively. Accordingly, firebugs with reduced expression of AKH receptors showed a significantly lower mortality (by 1.6 to 2.9-folds), and lower general metabolism after EPN + AKH treatments. In addition, EPN application increased Akh gene expression in the corpora cardiaca (1.6 times), AKH level in the corpora cardiaca (1.3 times) and haemolymph (1.7 times), and lipid and carbohydrate amounts in the haemolymph. Thus, the outcomes of the present study demonstrate involvement of AKH into the antistress reaction elicited by the nematobacterial infection. The exact mechanism by which AKH acts is unknown, but results suggested that the increase of metabolism and nutrient amounts in haemolymph might play a role. S využitím molekulárních metod byl zkoumán původ druhu Spergularia ×kurkae, domnělého tetraploidního křížence mezi diploidním středoevropským endemitem S. echinosperma a běžným tetraploidním druhem S. rubra. Sekvence regionu ITS (internal transcribed spacer) potvrdily hybridní původ S. ×kurkae jakožto křížence mezi S. echinosperma a S. rubra. Jelikož u téměř všech jedinců S. ×kurkae byla pozorována neúplná homogenizace rodičovských homeologů (incomplete concerted evolution), lze předpokládat, že S. ×kurkae je mladým taxonem. Je pravděpodobné, že tento taxon vznikl lidským přičiněním jakožto následek introdukce druhu S. rubra na obnažená rybniční dna. The origin of Spergularia ×kurkae, a presumed tetraploid hybrid between diploid central-European endemic S. echinosperma and its widespread tetraploid congener S. rubra, was investigated using molecular tools. Sequencing of the internal transcribed spacer (ITS) confirmed the hybrid origin of S. ×kurkae and the parentage. As incomplete concerted evolution of the ITS region was observed in nearly all individuals of S. ×kurkae, it is probable that S. ×kurkae is a young taxon. We speculate that it might have evolved as a result of human-mediated introduction of S. rubra into fishponds. The established fractionation scheme of Psenner and Pucsko with consecutive BD and NaOH steps was shown to be ineffective in distinguishing between the stable crystalline (hydr)oxides and their more active amorphous and poorly crystalline forms. These forms highly vary in their ability to sorb P and thus in their ecological significance. Dissolution kinetics of the Fe and Al minerals were performed using both the BD and NaOH extraction solutions. The results were applied to modify the original fractionation scheme for better quantification of P associated with active Al and Fe (hydr)oxides, and better estimation of the amounts of these Fe and Al forms in soils and sediments.
