Mladí lidé v politických stranách a participace mladých lidí na veřejném životě v ČR

2020 - Czech The master´s thesis deals with the topic of political participation of young people. It mainly focuses on young people who are politically involved in the South Bohemian Region. The thesis is divided into two parts. The theoretical part first introduces some of the selected definitions of political participation. I deal with possible forms of political participation: conventional and unconventional.The work also introduces the activities that young people, according to various researches, spend their free time. Subsequently, the relationship of young people, aged 18 to 35 years, to politics is described. Possible causes of young people's lack of interest in public events are also presented.In this part, attention is paid to active and passive voting. Subsequently, the theoretical part deals with selected determinants of political participation. The practical part presents the conclusions of the research of political participation of young people in the South Bohemian region. The aim of the research was to use interviews to find out what the experiences of young political candidates are. Discover, what motivates young people to take an interest in public affairs and what possible obstacles they have had to face. Diplomová práce se věnuje tématu politické participace mladých lidí. Především se zabývá mladými lidmi, kteří se politicky angažují v Jihočeském kraji. Práce je rozdělena na dvě části. V teoretické části jsou nejprve představeny některé z vybraných definic politické participace. Zabývám se možnými formami politické participace, a to jak konvenčními, tak nekonvenčními. Práce dále seznamuje s aktivitami, kterými podle různých výzkumů mladí lidé tráví volný čas. Následně je popsán vztah mladých (ve věku od 18 do 35 let) lidí k politice. Jsou představeny i možné příčiny nezájmu mladých lidí o veřejné dění. V této části je věnována pozornost aktivní i pasivní volební účasti. Následně se teoretická část věnuje vybraným determinantům politické participace. V praktické části jsou představeny závěry výzkumu politické participace mladých lidí v Jihočeském kraji. Cílem výzkumu bylo pomocí rozhovorů zjistit, jaké jsou zkušenosti mladých politických kandidátů. Objevit, co mladé lidi motivuje k zájmu o veřejné dění a jakým případným překážkám museli čelit. Systematika zelených kokálních řas tříd Chlorophyceae a Trebouxiophyceae

2020 - English Aim of the review part is to summarize a current situation in the systematics of the green coccal algae, which were traditionally assembled in only one order: Chlorococcales. Their distribution into the lower taxonomical unites (suborders, families, subfamilies, genera) was based on the classic morphological criteria as shape of the cell and characteristics of the colony. Introduction of molecular methods caused radical changes in our insight to the system of green (not only coccal) algae and green coccal algae were redistributed in two of newly described classes: Chlorophyceae a Trebouxiophyceae. Representatives of individual morphologically delimited families, subfamilies and even genera and species were commonly split in several lineages, often in both of mentioned classes. For the practical part, was chosen two problematical groups of green coccal algae: family Oocystaceae and family Scenedesmaceae - specifically its subfamily Crucigenioideae, which were revised using polyphasic approach. Based on the molecular phylogeny, relevance of some old traditional morphological traits was reevaluated and replaced by newly defined significant characteristics. Hmyzí anti-stresové reakce vůči přírodním toxinům

2020 - Czech The aim of this bachelor thesis was to characterize the effect of Bt (Bacillus thuringiensis) toxin on adipokinetic hormone (AKH) levels in the brain, on the activity of digestive enzymes (lipases, proteases, glucosideases, amylases) in the midgut and on nutrient levels (lipids, proteins, glycides) in the haemolymph of the firebug (Pyrrhocoris apterus). Bt toxin was applied by injection to the adult males 24 hours before monitoring of the above mentioned physiological characteristics. The results showed that Bt toxin reduced AKH level in the brain, modulated level of nutrients in haemolymph, but did not change activity of digestive enzymes in the midgut. Cílem této bakalářské práce bylo charakterizovat účinek Bt (Bacillus thuringiensis) toxinu na hladině adipokinetického hormonu (AKH) v mozku, na aktivitě trávicích enzymů (lipy, proteázy, glukosidázy, amylázy) ve středním střevě a na hladině živin (lipidy, glycidy) v hemolymfě ruměnice pospolné (Pyrrhocoris apterus). Bt toxin byl aplikován injekčně dospělým samcům 24 hodin před sledováním výše uvedených fyziologických vlastností. Výsledky ukázaly, že Bt toxin snižuje hladinu AKH v mozku, mění hladinu živin v hemolymfu, ale nemá vliv na aktivitu trávicích enzymů ve středním střevě. 2020 - Czech In the presented work, I describe the ability of Eurasian Magpie (Pica pica) to recognize predator species differing in the threat they represent. I presented stuffed dummies of northern goshawk (Accipiter gentilis, juvenile and adult female), common buzzard (Buteo buteo), common raven (Corvus corax) and common pheasant (Phasianus colchicus) to magpie pairs within the area of České Budějovice. The magpie behaviour was recorded and analysed, to see any dummy species related differences. V této práci se zabývám schopností straky obecné (Pica pica) rozpoznávat druhy predátorů, lišících se v hrozbě, kterou představují. Testoval jsem reakci párů strak na vycpané atrapy jestřába lesního (Accipiter gentilis, juvenilní a dospělé samice), káně lesní (Buteo buteo), krkavce velkého (Corvus corax) a bažanta obecného (Phasianus colchicus) v Českých Buděovicích. Chování strak bylo nahráno a analyzováno se snahou odhalit rozdíly závislé na druhu atrapy. Optimization of CRISPR/Cas9 technique for the Ixodes ticks genome editing

2020 - English CRISPR/Cas9 technique was used for genome editing of several arthropod species, but it has not yet been used on Ixodes spp. tick species. Therefore, in my work, I performed a bioinformatic analysis of the I. ricinus transcriptome and I. scapularis genome to reveal the sequence of the target gene for the CRISPR/Cas9-mediated knock-out. We prepared single guide RNA and mixed it with the Cas9 endonuclease to produce the Ribonucleoprotein complex and cleave the target sequence of the I. ricinus gene beta-galactoside alpha-2,6-sialyltransferase. Potravní chování skorce vodního (Cinclus cinclus) na tocích okresu Tachov

2020 - Czech In the first part of the thesis, characteristics (physical, environmntal, quantity and quality of prey) of favourable food patches and reference plots inside the Dipper´s territories were examined. In the second part, correlations of prey (quantity and quality) characteristics and physical-environmental characteristics were examined. In the third part, correlations between single types of observed Dipper behaviour were examined. In the fourth part, correlations between characteristics (physical, environmntal, quantity and quality of prey) of food patches and observed time of the Dipper´s foraging behaviour were examined. In the fifth part of the thesis, correlations between the territory characteristics (physical, environmntal, quantity and quality of prey) and body measurements of single birds were examined. V první části práce byly srovnány charakteristiky (fyzikální, environmentální, potravní nabídka) předpokládaných loveckých a kontrolních stanovišť v rámci teritoria skorce. Ve druhé části byly analyzovány vztahy mezi potravní nabídkou a ostatními charakteristikami teritoria. Ve třetí části byly analyzovány vztahy mezi jednotlivými typy sledovaného chování. Ve čtvrté části byly analyzovány vztahy mezi podílem času věnovaného loveckému chování a charakteristikami příslušného stanoviště. V poslední, páté části byly analyzovány vztahy mezi tělesnými rozměry odchycených ptáků a charakteristikami teritoria (fyzikálně-environmentální, potravní). 2020 - Czech This bachelor thesis sums up an overview of the current findings regarding floodplains and their usage. The pilot study focuses on the delimitation of floodplains of the right tributaries of the Vltava river on the territory of České Budějovice, alongside with the monitoring of the hydromorfological indicators of ecological guality of the Vrátecký stream. The thesis gives a description of the encounters between the functions and the current state of the floodplains' utilization. The thesis also contains a project proposal for terrain monitoring of the remaining tributaries of the Vltava river - the Dobrovodský stream, the Rudolfovský stream and the Hodějovický stream. Bakalářská práce shrnuje přehled současných poznatků o údolních nivách a jejich využívání. Pilotní studie se zabývá vymezením údolních niv pravostranných přítoků Vltavy na území Českých Budějovic a monitoringem hydromorfologických ukazatelů ekologické kvality Vráteckého potoka. V práci jsou popsány střety mezi funkcemi a současným stavem využívání. Součástí práce je návrh projektu na monitoring niv zbývajících přítoků Vltavy - Dobrovodského potoka, Rudolfovského potoka a Hodějovického potoka. Blood glucose concentration in Barn Swallow (\kur{Hirundo rustica}): sources of variability and association with fitness

2020 - English Glucose is an important fuel for intense activities of short duration, but in high concentration is reputed to be tissue-damaging at least in mammals. In birds, blood glucose concentration is naturally considerably higher than in other vertebrates of similar body mass. In this thesis, we focused on blood glucose concentration in the wild populations of the Barn Swallow (Hirundo rustica). Firstly, we investigated the level of repeatability of blood glucose concentration to find out if this trait is individual-specific and thus, it is subject to selection. Secondly, we examined which environmental and physiological variables explain variation in blood glucose level. Thirdly, we tested association of blood glucose level with fitness related traits, namely individual body mass and lifespan. Glukóza je jedním z hlavních zdrojů energie cirkulující v krvi obratlovců, který je využívaný pro krátkodobé intenzivní aktivity. Nicméně je známé, že v nadměrných koncentracích způsobuje u savců poškození tkání. U ptáků je koncentrace krevní glukózy výrazně vyšší než u ostatních skupin obratlovců srovnatelné velikosti. V této práci jsme se zaměřili na hladinu krevní glukózy u vlaštovky obecné (Hirundo rustica). V první kroku jsme se zabývali otázkou, jestli je hladina glukózy opakovatelným znakem a lze ji proto považovat za individuální znak, který může podléhat selekci. Následně jsme se pokoušeli identifikovat možné zdroje variability pocházející jak z vnějších, tak i vnitřních příčin. Tato práce se také zabývá souvislostí krevní glukózy s kondicí, konkrétně individuální hmotností a délkou dožití.
