Experiments such as antibiotic susceptibility test, PCR, growth curves and autoagglutination assays were included to best characterise the mentioned properties of the strains. Charakterizace klinických izolátů M. catarrhalis za použití molekulárně biologických metod a variačních metod identifikace bakterií na základě fenotypových odlišností. Mezi tyto metody patří např. test rezistence na antibiotika, PCR, test růstových křivek, rozbor schopnosti autoaglutinace a další. Tyto experimenty byly vybrány na zakládě pokrytí co největšího spektra vlastností, a tudíž co nejlépe tyto izoláty charakterizovat. Role of auxin-binding-proteins (ABP) in growth of corn seedlings mediated by potassium channel ZMK1

2009 - English The aim of this thesis is to collect current data about the topic: Role of auxin-binding proteins (ABP) in growth of corn seedlings mediated by potassium channel, ZMK1, and elaborate the diploma thesis. This thesis is composed of two main parts, theoretical part and experimental one. In the first part, we have focused on analysis of literature related to the topic of auxin-mediated growth responses of plants, auxin biosynthesis and transport, role and structure of auxin-binding proteins (ABPs), TIR1 signaling pathway, and role of potassium channels (especially ZMK1) in auxin-induced cell growth. In the second, experimental part we have focused on the investigation of the role of ABPs and inwardly rectifying potassium channel ZMK1 in auxin-induced cell growth. All experiments were performed on maize mutants abp1, abp4 and double mutant abp1/abp4 in corn (Zea mays L.) derived from near-isogenic line (WT). In these experiments, we have studied the responses of intact etiolated seedlings and excised mesocotyl segments to an inhibitor of potassium channels (tetraethylamonium chloride, TEA+). We also studied the expression of ZMK1 gene in mesocotyls of abp mutants and WT. Moreover, we have also tried to analyse the sequence of ABP4 promoter in two corn hybrids (307 and 3394). The experiments resulted in several interesting observations. We found that ABP1 and ABP4 mediate NAA-induced inhibition of coleoptile and mesocotyl elongation in etiolated corn seedlings. Our results also suggested that in auxin-induced inhibition of coleoptile and mesocotyl elongation of intact etiolated maize seedlings K+ channel ZMK1 is involved. Our molecular experiments also revealed that the mutations in ABP genes affect expression ZMK1 gene, and let to a proposal a working model of the interaction. Our results led to a general conclusion that in intact etiolated maize seedlings ABPs and ZMK1 cross-talk during auxin-induced growth inhibition. Cílem této práce bylo shromáždění aktuálních informací týkajících se tématu: Úloha auxin-binding proteinů (ABP) v regulaci růstu kukuřice zprostředkovaném draslíkovým kanálem ZMK1; a vypracování této diplomové práce. Tato práce se skládá ze dvou hlavních částí, teoretické a praktické. V první části jsme se zaměřili na analýzu literatury související s tématem růstových reakcí rostlin, zprostředkovaných auxinem, biosyntézy a transportu auxinu, role a struktury auxin-binding proteinů (ABPs), TIR1 signální dráhy a role draselných kanálů (zejména ZMK1) v buněčném růstu indukovaném auxinem. Ve druhé, experimentální části jsme se zaměřili na výzkum role ABP a vtokového draselného kanálu ZMK1 v buněčném růstu zprostředkovaném auxinem. Veškeré experimenty byly provedeny na mutantech kukuřice (Zea mays L.) abp1, abp4 a dvojitém mutantu abp1/abp4 odvozeném z téměř izogenní linie kukuřice (WT). V těchto experimentech jsme studovali reakce intaktních etiolizovaných rostlin a mezokotylových segmentů k inhibitoru draselných kanálů (tetraethylamonium chlorid, TEA+). Také jsme studovali expresi genu ZMK1 v mezokotylech abp mutantů a WT. Navíc jsme se pokusili o analýzu promotoru genu ABP4 ve dvou hybridech kukuřice (307 a 3394). Experimenty vedly k objevu několika zajímavých faktů. Zjistili jsme, že ABP1 a ABP4 zprostředkovávají inhibici prodlužování koleoptylu a mezokotylu rostlin kukuřice, způsobené NAA. Naše výsledky také naznačují, že auxinem indukovaná inhibice prodlužování koleoptylu a mezokotylu intaktních etiolizovaných rostlin se účastní draselný kanál ZMK1. Naše experimenty na molekulární úrovni také odhalili, že mutace v ABP genech ovlivňuje expresi genu ZMK1 a připouští tak návrh pracovního modelu interakce obou genů. Naše výsledky vedly k obecnému závěru, že v intaktních rostlinách kukuřice spolu ABP a ZMK1 spolupracují během inhibice růstu způsobené auxinem. The application of microcredit in Mexico

2009 - English Microfinance programs, providing financial services such as (micro)credit, insurance and saving to the poor, became a popular tool in the fight against the world poverty. The paper describes the development of this phenomenon and its impact in reducing the consequences of poverty in developing countries. Concretely, the work focuses on poverty related issues in Mexico. The paper brings description of various microfinance models used world-wide and shows examples of microfinance institutions operating in Mexico. It presents a strategy applied by a Mexican microfinance institution ADMIC Nacional and based on its experience and experience of its clients, it reflects on several aspects of microfinance culture in Yucatan. The work also identifies the challenges facing the current microfinance sector. It addresses especially the question of subsidization of microfinance programs which allows the programs to focus on poorer clients and provide them also with non-financial services, but, on the other hand, can constrain the growth and sustainability of the program. Mikrofinanční programy, poskytující finanční služby, jako například (mikro)půjčky, pojištění či spoření, chudým, se staly populárním nástrojem v boji proti světové chudobě. Práce popisuje vývoj tohoto fenoménu a jeho dopady na snižování následků chudoby v rozvojových zemích. Konkrétně se při tom zaměřuje na problematiku chudoby v Mexiku. Práce přináší popis různých modelů mikropůjček užívaných ve světě a ukazuje příklady mikrofinančních institucí v Mexiku. Představuje strategii užívanou organizací ADMIC Nacional a na základě zkušeností této organizace a jejich klientů se zamýšlí nad několika aspekty mikrofinanční kultury na Yucatánu. Práce také identifikuje výzvy, jimž čelí současný sektor mikrofinancí. Především se zabývá otázkou dotování mikrofinančních prgramů, které programům umožňuje soustředit se na chudší klienty a poskytovat jim také jiné než finanční služby, na druhou stranu však může programy omezovat v jejich růstu a dosažení udržitelnosti. It seek the similarities but also dissimilarities between the two languages. The practical part focuses on teaching and learning the passive voice at Czech lower secondary schools. The tests reveal what pupils know but also what they have problems with. The questionnaires try to find out the teachers´ opinions about teaching the passive voice. The Competition of the Definite Article and Possessive Pronous

2008 - English The diploma thesis examines the competition between the definite article and possessive pronouns. The theoretical part concernes with the study of literature, mainly the grammar books by Quirk, Poldauf and Dušková. However, this phenomenon is not thoroughly described, so the focus of this thesis rests on its own research. British National Corpus, which is a database containing 100 million word collection of samples of written and spoken language from a wide range of sources, is used in the practical part. The thesis works especially with Quirk{\crq}s examples and these are analysed in a detailed manner. It is examined in which cases the competition between the definite article and possessive pronouns factually exists, what influences the preference of the article or possessive or if it is possible to use the article or possessive pronoun without any change in the semantic structure (and if there is a semantic change, so what kind of change it is). The phenomenon is thus researched from the point of view of semantics, interchangeability, grammaticalization and stylistics. The particular examples relate to the semantic fields concerning parts of the body, parts of the clothing or family members. Diplomová práce zkoumá konkurenci určitého členu a přivlastňovacího zájmena. V teoretické části se opírá o studium literatury (zejména gramatik od autorů Quirk, Poldauf, Dušková). Protože tato problematika není detailněji zpracována, hlavní centrum práce spočívá ve vlastním výzkumu. V praktické části je používán Britský národní korpus, což je databáze obsahující 100 milionů slov mluveného i psaného jazyka z širokého spektra zdrojů. Zde se pracuje převážne s Quirkovými příklady, které jsou podrobeny detailní analýze. Zkoumá se, ve kterých případech existuje konkurence určitého členu a přivlastňovacího zájmena, co ovlivňuje preferenci jednoho nebo druhého nebo jestli je možné použít jak určitý člen, tak přivlastňovací zájmeno, aniž by došlo k jakékoliv sémantické změně (pokud k sémantickému posunu dojde, tak k jakému). Tento fenomén se tedy zkoumá z hlediska sémantiky, ale také gramatikalizace i stylistiky. V konkrétních příkladech se nejčastěji objevují části těla, oblečení nebo členové rodiny.
