This study concerns clarification of the origin of infections of arvicoline rodents with Isospora spp. based on three different approaches: phylogenetic analyses of three genes (18S rRNA, COI and COIII), morphological and morphometrical analyses, and experimental infection. Field collections, parasitological examinations of samples, microscopy, DNA extraction, PCR, and computational analyses were employed during the course of this study.

Proteins involved in the tetrapyrrole pathway in Synechocystis sp. PCC 6803 and their localization in the proximity of PSII biogenesis

The goal of the thesis was to enhance our understanding of the tetrapyrrole pathway in cyanobacteria by a study of selected proteins involved in the pathway. During the project I have revealed functional connection between protoporphyrinogen IX oxidase HemJ and preceding enzyme in the pathway by complementation of protoporphyrinogen IX oxidase deletion mutant by its analog HemG from Escherichia coli. Heme b was identified as a cofactor of HemJ. Another protein deeply influencing tetrapyrrole accumulation, BtpA was found to form a complex with GluTR, the enzyme at the beginning of the tetrapyrrole pathway. Lastly, CurT protein, the component of the structures anticipated to function in PSII assembly and/or repair localized at plasma/thylakoid membrane interface, was co isolated with the enzymes of the tetrapyrrole pathway suggesting that the specific CurT containing membranes could be a place of both photosystem II assembly/repair and chlorophyll delivery.

The aim of this thesis is to investigate community assemblages of fruit and seed feeding insects across three distinct biogeographic ForestGEO plots of Baro Colorado in Panama, Khao Chong in Thailand and Wanang in Papua New Guinea. It studies fruit and seed syndromes from which insects were reared per plant species across this rain forest plots, and describe host specialization in fruit and seed feeding insects. More specially, to compare seed feeding insects and their rate of seed attack among different insect groups are consistent across rainforest sites. Further it use plant phylogeny to explore plant floristic diversity, and explain ecological role of seed insect specialization in regulating plant species dynamic in maintaining high plant diversity in tropical rainforest regions.

Biology of the Mitochondrial Contact Site and Cristae Organizing System in Trypanosomatids

This study describes Mitochondrial Contact Site and Cristae Organizing System (MICOS) in unicellular parasitic protist Trypanosoma brucei. This is the first study of MICOS outside of canonical model organisms belonging to the supergroup Opisthokonta. Nine subunits of the complex were purified and characterized. Their role in cristae biogenesis and interaction with other mitochondrial protein complexes was demonstrated. Further investigation revealed a hierarchical organization of T. brucei MICOS and the existence of two subcomplexes. One of MICOS subunits, TbMic20, facilitates protein import into mitochondrial intermembrane space, which is one of the novel features of T. brucei MICOS. The role of TbMic20 in oxidative protein folding was demonstrated and the mechanism of this process in T. brucei was addressed.

LANGUAGE CHANGE IN TERMS OF GENDER IN THE ERA OF EARLY MODERN ENGLISH

Gender v jazyce je jedno z velmi diskutovaných témat sociolingvistických studií. Podobně kontroverzní je i téma jazykové změny, která stále bývá považována za úpadek jazyka namísto jeho vývoje. Tato bakalářská práce se bude zabývat vztahem mezi těmito okruhy. Zároveň se bude zaměřovat i na vůdce jazykových změn. Na základě důkladného prostudování mnoha sociolingvistických studií byla stanovena hypotéza, která ukazuje na ženy, jakožto vůdce jazykových změn. Verifikace hypotézy zahrnuje vybrané jazykové změny z období rané moderní angličtiny, na kterých bude analyzováno jejich rozšíření s ohledem na iniciátory těchto změn. Výzkum bude prováděn na základě osobní korespondence z období rané moderní angličtiny. Language and gender has always been a major subject for sociolinguistic discussion. Moreover, the same applies to language change, which is still by some considered to be rather a decay in language than its progress. This thesis aims to investigate the relation between these two areas. Furthermore, it tries to establish the leaders of language changes with regard to gender. Based on the analysis of many sociolinguistic studies, this paper comes up with the hypothesis that women are the leaders of language changes. The process of verifying the hypothesis comprises few selected examples of language changes in the period of Early Modern English, on which is analysed their spread and their initiators. The background for this thesis are the personal letters from the period of Early Modern English.

Neglected aspects of bark beetle (Coleoptera: Scolytinae) ecophysiology

The thesis describes several unknown aspects of the spruce bark beetle, Ips typographus (L.), and the double-spined bark beetle, Ips duplicatus (Sahlberg.), life-cycles and ecophysiology. The first study focuses on I. typographus and its dispersal under epidemic conditions in the National park Šumava and introduces a novel method of fluorescent marking and detection of captured specimens. The second study is focused on ability of I. typographus to establish so-called sister-broods, i.e. re-emergence of females that are capable to continue egg laying without a need to mate again. The importance of sister-broods becomes apparent mainly in recent hot and dry vegetation seasons, which is demonstrated by a comparison of recent and historical data. The third study focuses on temperature-dependent development of I. duplicatus under laboratory conditions by the means of sandwich method. Altogether, the studies underline practical importance of ecophysiological studies on bark beetles as one of the tools for their effective management. Cílem diplomové práce je vytvořit kvalitativní výzkum za účelem analýzy zkušeností účastníků projektu NetMe-In. Předložená práce se skládá z teoretické části, popisu projektu a konkrétního týmu, metodologie výzkumu, empirické části a diskuse. Nejprve je představena teoretická část, která se věnuje čtyřem hlavním tématům: projekt, tým, interkulturalita a mnohojazyčnost. Dále je podrobně vysvětlen koncept projektu NetMe-In a jsou představeni jeho partneři. Třetí část se věnuje metodologii výzkumu, včetně popisu autorovy metody, představení záznamového zařízení, analýzy dotazovaných účastníků, představení průběhu interview a transkripčnímu přepisu rozhovorů. Čtvrtá část diplomové práce se zabývá analýzou a interpretací získaných rozhovorů. Závěrečná část je zaměřena na diskuzi, autorovu kritickou reflexi kvalitativního výzkumu, včetně komentářů k autorově metodologii a výsledkům výzkumu. The purpose of this work is to carry out qualitative research in order to obtain an in-depth understanding of the NetMe-In project participants´ experiences. The Master thesis is made up of theoretical part, project and team description, research methodology, empirical part and discussion. Firstly, the theoretical part is focused on four main topics: project, team, interculturality and multilingualism. Secondly, a detailed description of the NetMe-In project itself as well as an introduction to all project partners is provided. Thirdly, the research methodology is presented including the author's research method, the recording equipment, the description of interviewees, the interview settings and the description of the interview transcription. Fourthly, the empirical part puts emphasis on analysis and interpretation of the results of the author's interviews. Lastly, the discussion is concentrated on the author's critical reflection of the qualitative research including comments on the author's methodology and the research findings.

Tato diplomová práce se zabývá hrdiny románů J.K. Rowlingové, zahrnující fantasy sérii Harry Potter, detektivní romány řady Cormoran Strike a román pro dospělé čtenáře Prázdné místo. Hlavním cílem této práce je popsat archetypy hrdinů vyskytujících se ve zkoumaných příbězích a také vylíčit typy zkoušek, kterými tito hrdinové na své cestě musí projít. Následující část práce se pokouší vypozorovat převažující typy archetypů hrdiny a rovněž zkoušek s ohledem na měnící se literární žánry. Praktická část poukazuje na podobnosti mezi všemi romány od J.K. Rowlingové s hlavním zaměřením na mateřské a otcovské postavy a jejich opakující se rysy. This diploma thesis deals with the heroes of J.K. Rowling´s novels, including her Harry Potter fantasy series, her Cormoran Strike detective series and her adult novel The Casual Vacancy. The aim of the thesis is to describe the archetypes of the heroes appearing in the stories and also to depict the types of the trials these heroes have to pass on their journey. In the following part the thesis observes the predominant type of the archetypes and the trials with respect to changing literary genres. The practical part points out the similarities between all Rowling´s novels with the main focus on mother and father figures and their repetitive characteristic features.

Cílem bakalářské práce: "Variabilita anglického slovního přízvuku" je popsat základní problematiku spojenou s umístěním přízvuku v angličtině se zaměřením na lexikální jednotky, které mají více možností umístění přízvuku, aniž by se měnil jejich význam. Praktická část práce se pak zabývá sběrem lexikálních jednotek s variabilním přízvukem, na jejímž konci bude uveden abecední seznam variujících slov spolu s transkripčním přepisem jednotlivých přízvukových variant. The goal of the bachelor thesis: "Lexical stress variability in English" is to describe basic principles of stress assignment in English with a focus on lexical units that have more realisations of stress placement without changing the meaning of the word. The practical part concentrates on collecting lexical items with variable stress. There is going to be an alphabetical list of stress-divergent words along with phonetic transcription of stress variations at the end of the work.
