Preliminary analysis of reproductive success in the Northern chamois (Rupicapra rupicapra) from NP Grand Paradiso

Paternity analysis was performed on a study population of chamois (Rupicapra rupicapra) from 107 feaces samles. DNA was extracted from samples using commercial kits (Qiagen). Based on analysis of 17 microsatellite loci was found that a 6 different males sired a total of 9 kids. Male M13_16 sired more than half the kids. Each of the five other males sired always only one kid.\n Pro zjištění paternity a příbuzenských vztahů ve vybrané populaci kamzíka horského (Rupicapra rupicapra) byly provedeny analýzy ze vzorků trusu 107 jedinců. DNA byla ze vzorků izolována pomocí komerčních extrakčních kitů (Qiagen). Na základě analýz 17 mikrosatelitových lokusů bylo zjištěno, že 6 různých samců zplodilo celkem 9 mláďat. Téměř polovina potomků měla za otce jednoho samce M13_16. Každý z ostatních pěti samců zplodil vždy jen jedno mládě. Seasonal dynamics of ergasilosis in reservoir fish

We evaluated ergasilosis seasonal dynamics at two reservoirs (Hubenov, Korycany) in the Morava River Basin (Czech Republic). Samples of fish were obtained at monthly intervals between April and October 2014. In total, 189 fish of 11 species were caught using electrofishing and seine nets. Highest overall values for E. sieboldi infection intensity and abundance were recorded at Hubenov in June and September. Prevalence ranged between 80 and 100%. Two highest peaks of infection intensity and abundance were recorded at Korycany in May and the second around September and October, prevalence ranging between 33 and 100%. As the nauplius and copepod stages of arthropods form part of the zooplankton assemblage, they will be found at highest numbers in reservoirs with low predatory pressure, i.e. where predatory fish suppress zooplanktonophagic fish species. Toxic effect of fluorescence pigment on zebra fish (Danio rerio)

The aim of the study was to determine the toxic effects of organic pigments Alizarin Red S and Alizarin Complexone on zebra fish (Danio rerio). For short-term acute toxicity tests on zebra fish concentration of 150; 300 and 600 mg/L were chosen for both dyes. Toxic effect of dyes was observed even in the variant with 10 g/L of sodium chloride. Addition of sodium chloride increases the deposition of dyes in the bone structures of the fish. LC50 values were analyzed graphically by using probit analysis. There was no mortality during the acute toxicity test with Alizarin Red S even at the highest concentration. Toxicity value (72hLC50) for zebra fish with a combination of Alizarin Red S + 10 g/L of sodium chloride is 546.42 mg/L. Mortality for Alizarin Complexone was 100% in 24 hours at\nconcentrations of 300 and 600 mg/L, with concentrations of 150 mg/L there was no mortality. In Alizarin Complexone supplemented with 10 g/L of sodium chloride was 100% mortality at all\nconcentrations up to 72 hours. The project was focused on the survey and evaluation of biomanipulation effects performed on the drinking water reservoir Hamry and ichthyological survey of drinking water reservoirs Souš and Josefův Důl. The evaluation of current composition of fish assemblages in the area of the future Pěčín reservoir. Projekt byl přednostně zaměřen na průzkum a vyhodnocení účinku biomanipulace na vodárenské nádrži Hamry a ichtyologický průzkum vodárenských nádrží Souš a Josefův Důl. Terénním šetřením bylo zahájeno vyhodnocení současného stavu rybích společenstev v zátopě připravované nádrže Pěčín. To determine the present state a questionnaire was prepared with 17 questions mapping the following topics: control and knowledge of the lineage bred, outline of the reproduction technologies and alternative genetically improved techniques used, sign and type of the selection made, type of the breeding method involved, knowledge and degree of utilization of genetic tools, knowledge of the latest breeding programs used abroad. Pro zjištění současného stavu byl připraven dotazník s 17 otázkami mapujícími okruhy: kontrola a znalost chované linie, nástin používaných reprodukčních technologií a alternativních geneticky vylepšovaných technik, znak a typ prováděna selekce, typ používané šlechtitelské metody, znalost a míra používání genetických nástrojů, informovanost o nejnovějších chovných programech v zahraničí. This study, which has been running since 2008, examines the use of the Hamry Water Reservoir as a model for fish community biomanipulation studies with the aim of improving water quality. Of primary interest to the Reservoir administrators, the Elbe River Basin authority s.p., is the maintenance of high water quality for drinking purposes, thereby preventing conflict of interest with multiple stakeholders. In addition, the study aims to determine whether biomanipulation of fish communities can be undertaken effectively in a reservoir. Cílem celé studie, která probíhá již od roku 2008, je využití VN Hamry jako modelového objektu pro biomanipulační zásahy v rámci zlepšení kvality vody v nádrži. Prvotním zájmem správce VN, podniku Povodí Labe, s.p., je vysoká kvalita vody pro pitné účely, takže zde nedochází ke konfliktu zájmů více zainteresovaných stran. Cílem bylo rovněž zjistit, zda je vůbec reálné v podmínkách VN provádět efektivní biomanipulace s rybím společenstvem. In 2015, we surveyed the newly established fish community in order to acquire information for future fisheries management.
