Evaluation of proposed revitalization of the river Ploučnice close to Mimoň in terms of remobilization of historical contamination

The aim of the study was to estimate the uranium contamination in the alluvial sediments of original river beds of the Ploučnice river between 72-73 river kilometer on a left bank, near Mimoň town, and evaluate if the contamination should be included into preparation of revitalization projects. The study of contamination was started by mapping the gamma activity of the surface of the floodplain. Based on the resulting map the sediment samples were taken by hand earth drill from the former floodplain and channels abandoned in the 1980s by regulations. The samples were analysed by laboratory X-ray fluorescence (XRF). This step was followed by subsequent mapping by handheld XRF to specify the contamination pattern in the floodplain in more details. The evolution of watershed as well as the exact terrain model were processed by GIS program. Two major contaminated areas have been found, and further sampling was conducted. This enabled us to reconstruct the historical development of contamination at exceptional time scale resolution and describe precisely the evolution from the 19th century to the time of the regulation of that river reach. It has also been shown, that any revitalization in the near future would be probably thwarted due to extremely fast river changes which would cause an immediate re-introduction of the historical contamination into the fluvial system. Cílem této studie je zhodnotit historickou kontaminaci uranem v nivních sedimentech a původních korytech Ploučnice mezi 72. až 73. říčním kilometrem Ploučnice, v nivě na levém břehu řeky, cca 800m pod městem Mimoň a zdůvodnit, proč by mělo posouzení historické zátěže být zařazeno do procesu příprav revitalizačních prací. Bylo provedeno mapování gama aktivity na povrchu nivy, na jehož základě byly ručním žlábkovým vrtákem odebrány hloubkové profily nivních sedimentů i vzorky ze starých koryt odstavených regulacemi v 80. letech, které byly analyzovány rentgenovou fluorescenční spektrometrií (XRF). Následně byla provedena další mapování ručním XRF spektrometrem s cílem upřesnit rozložení uranu v jednom z nejvíc kontaminovaných míst v nivě. Vývoj vodního toku včetně studia přesného modelu terénu byl zpracován pomocí GIS. Analýza hloubkových profilů kontaminantů v nivě umožnila rekonstruovat vývoj historické kontaminace s výjimečným časovým rozlišením, představujícím zatím nejpodrobnější záznam vývoje znečištění od konce 19. století do regulace toku. Bylo prokázáno, že v minulosti docházelo k takovým rychlým bočním posunům meandrujícího toku, že by v důsledku případné revitalizace v blízké budoucnosti pravděpodobně došlo k remobilizaci historické kontaminace zpět do říčního systému. Detection of Dasatinib using Pre-reaction with Copper and Mass Spectrometry

The determination of dasatinib (DAS) was provided using front-end electrochemical separation with mass detection. The electrochemical separation is based on the reaction of DAS with copper solid electrode. The affinity of DAS to copper is supported by redox reaction yealding to DAS fragmentation. Stanovení dasatinibu (DAS) bylo provedeno pomocí elektrochemické separace následované hmotnostně spektrometrickou detekcí. Elektrochemická separace je založena na reakci DAS s pevnou měděnou elektrodou. Afinita DAS k mědi je podpořena redoxní reakcí vedoucí k fragmentaci DAS. Analysis of low abundant lipids in vernix caseosa using chromatographic methods and mass spectrometry

The aim of this work is to develop a separation method for characterization of unknown lipids in vernix caseosa. We focused on very low abundant lipids which have not been previously described in vernix caseosa. The total lipid extract was fractionated using low pressure normal phase liquid chromatography to obtain sub-fractions. The sub-fractions were further separated in high performance liquid chromatography mode to isolate classes of lipids. This procedure enabled us to characterise lipid classes by mass spectrometry and identify one of the class as acyl-ceramides with non-hydroxylated fatty acids. Thirty compounds were investigated in order to detect their repellent and antifeedant activity. Chiroptical spectroscopy as a sensitive tool for the conformational analysis of antimicrobial peptides

We tested the utilization of electronic and vibrational circular dichroism for the conformational study of antimicrobial peptides on two examples: melectin and antapin in different solutions of liposomes and micelles mimicking bacterial or eukaryotic membranes. The results show significant content of the alpha-helical conformation in an environment mimicking the bacterial membrane, while it was considerably low in the eukaryotic membrane-mimicking environment. Capillary electrophoretic study of solvent effects on complexation of dibenzo-18-crown-6 with potassium ions

Affinity capillary electrophoresis was applied to investigation of solvent effects on stability constant of dibenzo-18-crown-6 complex with potassium ion in mixed hydro-organic solvents at different water/methanol, water/ethanol and water/propan-2-ol ratios. The apparent stability constants of the complex were found to be strongly decreasing with increasing molar fraction of water in the mixed solvents.
