Cross-amplification of microsatellites on European Ranunculus sect. Batrachium species was tested using primers that were designed for R. bungei. In this thesis, environmental and clinical Streptomyces strains were used to test the hypothesis if isolates from different isolation sources vary in frequency of beta-hemolytic activity. Moreover, their interaction with two human pathogens Pseudomonas aeruginosa and Staphylococcus aureus was tested in parallel and T-type co-cultivations. For this, the responses (beta-hemolytic activity, growth, etc.) of both streptomycetes as well as model pathogens were evaluated. Furthermore, the frequency of observed interaction patterns with tested pathogens were compared between environmental and clinical Streptomyces strains as such study is lacking. Recombinant proteins derived from Bombyx mori Ser-2 gene and Drosophila melanogaster Sgs3 gene were expressed in bacterial expression systems, purified and analyzed using different molecular biology techniques. The pure proteins were tested for cell adhesive properties by coating hydrophobic surfaces with the proteins and monitoring the growth of adherent cells in vitro. By showing adequate expression and resultant yield, as well as the ability to aid cell adhesion, the adhesive proteins present great potential for future biomaterials. Recombinant proteins derived from Bombyx mori Ser-2 gene and Drosophila melanogaster Sgs3 gene were expressed in bacterial expression systems, purified and analyzed using different molecular biology techniques. Functional characterization of succinyl-CoA synthetase in the bloodstream form of Trypanosoma brucei

During the infectious bloodstream stage (BF), the human pathogen Trypanosoma brucei lacks a canonical cytochrome-mediated respiratory chain. Therefore, this form of the parasite depends on the reverse activity of the FoF1-ATP synthase to hydrolyze ATP and pump protons into the mitochondrial intermembrane space to maintain the essential membrane potential. The dogma also states that the sole source of organellar ATP is the mitochondrial ADP/ATP carrier, which imports glycolytically derived ATP from the cytosol. In this study, we explore the possibility that the BF mitochondrion is able to contribute to its own pool of consumable ATP by utilizing an active succinyl-CoA synthase to generate ATP via substrate phosphorylation. Trehalose as an important energy source for immune cells in Drosophila melanogaster

The aim of this thesis was the identification of the change of trehalase gene expression in hemocytes of Drosophila melanogaster in uninfected and infected larvae using parasitoid wasp infection by Leptopilina boulardi. Moreover, the energy supply was shut down by deactivating the trehalose transporter and measuring the lamellocyte production, again in uninfected and infected larva to test the role of trehalose for the selfish immune system. Secondary metabolites, produced by various microbes, are used since decades for treatment of diseases. The bacterial genus Kutzneria is known for synthesizing compounds with antifungal activity, which might be used against pathogenic fungi, causing agricultural relevant diseases. This thesis deals with the genome analysis of the strain Kutzneria sp. BCCO 10_1627 for prediction of secondary metabolite gene cluster. Moreover, the antifungal activity of the strain was determined experimentally. Secondary metabolites, produced by various microbes, are used since decades for treatment of diseases. Tato bakalářská práce se zabývá vyučovacími metodami a tím jako jsou aplikovány v praxi. Zaměřuje se především na jejich vývoj a popisuje techniky a principy, které jsou pro ně charakteristické. Praktická část vychází z poznatků získaných při pozorování hodin v jazykových školách a zkušenosti jedné účastnice, která absolvovala sugestopedický kurz. V každém kurzu jsou pozorovány stejné aspekty, které hrají důležitou roli, co se týče úspěšnosti přístupu ve vyučování. Tato práce si klade za cíl poukázat na silné a slabé stránky jednotlivých metod a porovnat výsledky z pozorovacích hodin. Abstract This thesis is concerned with language teaching methods and how they there are applied in practise. It focuses mainly on their development and description of techniques and principles that are characteristic for these methods. The practical part is patterned on findings gained from observation of classes in different language schools and also on experience of one student who attended the suggestopedic course. In each course the same aspects are observed, which play an important role in a success rate of an approach to teaching. The thesis sets out to show the advantages and disadvantages of particular methods and compared the results of classroom observation.
