The study aims to develop a system for detecting diabetic retinopathy using deep learning. In this study I have explored transfer learning with four distinct models and addressed the issue of an unbalanced dataset with oversampling. The final experiment achieved a significant improvement in accuracy and quadratic kappa score. The study highlights the potential of deep learning and the importance of addressing dataset imbalances for accurate results. Cílem této práce je analýza společenského postavení hlavních postav v románech Jane Austenové, konkrétně Pýchy a Předsudku a Emmy. Bakalářská práce je rozdělena do čtyř kapitol. První se zabývá autorkou Jane Austenovou, jejím životem a tvorbou a následně autory, jež Austenovou ovlivnili při jejím psaní. Druhá kapitola nám přibližuje dobu, kdy autorka žila, tedy Anglii 19. století. Době se přezdívalo Regentská Anglie a přiblížíme si zde roli a rozdíly v životě žen a mužů a ekonomickou situaci, jak obecně v 19. století, tak v románech Jane Austenové. Budou uvedeny příklady, kolik vydělávali hlavní hrdinové. Třetí část už se soustředí na samotnou hlavní postavu románu Emma. Popisuje rysy, kterými se Emma liší od ostatních hrdinek ve zbytku Austenových románů. Zároveň projednává její postavení v kruhu společnosti, ve kterém se vyskytuje. Druhá podkapitola odkrývá vztah s nejbližšími postavami, se kterými Emma přišla do kontaktu. Poslední kapitola Elizabeth Bennetová má trochu jinou strukturu než předchozí, i přes to že cílem práce je hlavní hrdinky vrhnou na stejné světlo. Vymezuje limity Bennetovy rodiny, které souvisí s jejich příjmem, a tudíž limituje jejich společenské postavení ve společnosti. V poslední kapitole je zahrnuta i Elizabetina ženskost. Na závěr práce porovnávám naše dvě hrdinky, konkrétně jejich osobnost, pohled na manželství, rozvoj charakteru a sociální postavení. This thesis aims to analyze the social status of the main characters in Jane Austen's novels, specifically Pride and Prejudice and Emma. The bachelor thesis is divided into four chapters. The first deals with the author Jane Austen, her life and work, and then focus on the authors who influenced Austen in her writing. The second chapter brings us closer to the time when the author lived in England in the 19th century. The era was called the Regency Era and we going to look at the role and differences in the lives of women and men and the economic situation, both in general in the 19th century and in Jane Austen's novels. Examples of how much the main male characters earned going to be given. The third part finally focuses on the main character of the novel, Emma. It describes the features that make Emma different from the other heroines in the rest of Austen's novels, while also discussing her position in the circle of society in which she existed. The second sub-chapter reveals the relationship with the closest characters with whom Emma came into contact. The last chapter of Elizabeth Bennet has a slightly different structure than the previous one, even though the work tries to cast the main characters in the same light. It defines the limits of the Bennet family, which is related to their income and therefore limits their social status in society. Elizabeth's womanhood is also included in the last chapter. At the end of the work, I compare our two heroines, specifically their personalities, views on marriage, character development, and social status.

The Reception of Harriet Jacobs' Incidents in the Life of a Slave Girl

Cílem této bakalářské práce je zaměřit se na problematiku recepce díla Incidents in the Life of a Slave Girl od afroamerické autorky Harriet Jacobs. Ačkoli je tato autobiografie, poprvé vydaná počátkem roku 1861, dnes známá jako jedno z nejpřednějších děl tzv. slave narratives, vyprávění otroků, jeho autenticita a autorství byly zpochybňovány po mnoho dekád, a teprve koncem 20. století byla kniha opět uznána jako hodnotné a pravdivé osobní svědectví toho, co pro afroamerické ženy znamenalo otroctví ve Spojených státech před občanskou válkou. Tato práce se tedy soustředí jak na recepci dobovou, s přihlédnutím především k historickým okolnostem abolicionismu a začátku občanské války, tak i na recepci současnou, zejména na kritické ohlasy 20. století a následnou kanonizaci díla. Klíčovým faktorem této analýzy bude mnohonásobná diskriminace, jež ovlivňovala život i práci Harriet Jacobs jakožto sexuálně zneužívané ženy afroamerického původu. The aim of this thesis is to explore the problematics of the reception of Harriet Jacobs' Incidents in the Life of a Slave Girl. Though this autobiography, first published at the beginning of 1861, is today known as one of the most major works of the genre of slave narratives, its authenticity and authorship have been questioned and denied for several decades, and only by the end of the 20th century has the narrative been recognized as a valuable and truthful personal testimony of what slavery meant for African American women in antebellum America. This thesis then focuses on both the contemporary reception, mainly with regard to the historical circumstances of abolitionism and the upcoming Civil War, and a more recent one, predominantly on 20th-century criticism and the subsequent canonization of the narrative. The key factor of this analysis will be the multiple discrimination which affected the life and work of Harriet Jacobs as a sexually abused woman of African American descent.

Role of the serotonin receptor type 7 (5-HT7) in autoimmune disease

Serotonin receptor type 7 (5-HT7) displays immunomodulatory functions and is overexpressed in individuals with Crohn's disease. Here, we set out to determine 5-HT7 mode of interaction with its cognate proteins Gs and G12. A unique type of coupling, termed 'inverse coupling' is known to occur between the 5-HT7 receptor and the Gs protein. The aim of our research was to determine whether G12 can also undergo this kind of coupling. We found that the G12 protein has a surprisingly lower mobility than Gs implying a specific interaction with a membrane domain or another membrane-localized protein. For both G proteins, we were unable to detect their inverse coupling with the 5-HT7 receptor at room temperature. Our findings indicate peculiar differences in G protein mobility and emphasize the importance of temperature for studies of interactions between G proteins and G protein-coupled receptors. Tato práce zkoumá vědecký základ agroekologických praktik, metod a struktur doporu-čené permakulturou, jak uvedené na dotazník České permakulturní asociace s účelem kvalifikovat permakulturní projekty jako takové čí jako projekty s permakulturními prv-ky. Pro praktiky, metody a struktury uvedené byla provedená literární rešerše na Web of Science. Přes tuto rešerši bylo možné identifikovat různé publikace potvrzující vhod-nost těchto doporučených praktik, i když pro některé z nich, zejména aplikace průmys-lových hnojiv a bezorebné systémy, vědecké výsledky naznačí pozitivní vlivy, dokud správně aplikované. Dále práce s tímto dotazníkem sugeruje, že permakultura není prostě agroekologická hnutí, ale její dnešní ohnisko je širší, hledající udržitelná řešení v mnoho aspektech života, včetně ubytování, získávání energie a vody a také socio-ekonomických aktivit lokálních a regionálních komunit. This thesis attempts to verify whether agroecological practices recommended by per-maculture are scientifically founded. For this purpose, a questionnaire used by the Czech permaculture association (Permakultura CS) to qualify projects as permaculture showcase projects or projects with permacultural elements, was used, and for the listed practices, methods and structures a literature search on Web of Science was per-formed. By the literature search we could identify several publications confirming the suitability of the recommended practices, although for some of them, in particular the avoidance of tillage as well as industrial fertilizers, scientific evidence suggests that, as long as correctly applied, these may even have a positive impact. Furthermore, the re-view of the questionnaire confirms that permaculture is not per se an agroecological movement, but its focus is nowadays broader, searching for sustainability solutions in most aspects of life, including housing, energy and water sourcing, as well as in socio-economic aspects of local and regional communities.

This study aims to explore the principle of the interdependence and mutual influence of the Dark web and cryptocurrency. Specifically, it explores the terms "the Dark web" and "cryptocurrency", as well as distinguishes cryptocurrency from normal currency. Furthermore, it establishes a connection between the Dark web and cryptocurrency. In order to reach the aim of this study, multiple sources of literature had been analyzed, including books and online articles. The analysis of literature shows that there is a strong correlation between the existence of the Dark web and demand on cryptocurrency. Thus, the results of this paper conclude that the Dark web and cryptocurrency exist in symbiosis - cryptocurrency allows the Dark web to exist for its anonymity and impunity, while the Dark web guarantees demand on cryptocurrency. Účinnost mykoparazitických a entomopatogenních hub proti houbovým původcům onemocnění rostlin

Entomopatogenní a mykoparazitické houby mají potenciál být využívány na místní i mezinárodní úrovni jako účinná a ekologicky šetrná strategie boje proti škůdcům a chorobám rostlin způsobeným houbovými patogeny. V bakalářské práci byla tes-tována účinnost užitečných hub v laboratorních podmínkách. Metodika zahrnovala izolaci a popis užitečných druhů hub a následné testování užitečných hub s poten-cionálními mykoparazitickými a antagonistickými účinky. Účinnost hub byla tes-tována na patogenech, které byly převážně odizolovány z osiva nebo nemocných ros-tlin luskovin. V rámci pokusů bylo zjištěno, že všechny kmeny Trichoderma izolo-vané z půd ČR prokázaly mykoparazitický efekt na vybrané patogeny. Nicméně, účinnost kmenů závisela na druhu houbového patogenu. Hodnocena byla i produkce spor kmenů Trichoderma jak v kontrolní variantě, tak i ve vzájemné kombinaci s patogeny. Výsledky opět prokázaly rozdíly v produkci spor u jednotlivých kmenů Trichoderma. Zároveň byl hodnocen mykoparazitický účinek entomopatogenních hub Isaria fumosorosea a Akanthomyces attenuatus proti patogenu Botrytis cinerea. Výsledky ukázaly, že ačkoli I. fumosorosea a A. attenuatus vykazovaly významný mykoparazitický účinek proti Botrytis cinerea, nejvyšší účinnost proti patogenu prokázala mykoparazitická houba T.virens. Zjištění naznačila, že jak mykoparazitické, tak entomopatogenní houby jsou udržitelnou alternativou umělých pesticidů v boji proti houbovým původcům chorob rostlin. Entomopathogenic and mycoparasitic fungi have the potential to be used both locally and internationally as an effective and environmentally friendly strategy for the man-agement of pests and plant diseases caused by fungal pathogens. In this bachelor thesis, the efficacy of beneficial fungi was tested under laboratory conditions. The methodology involved isolation and description of useful fungal species and subse-quent testing of useful fungi with potential mycoparasitic and antagonistic effects. The efficacy of the fungi was tested on pathogens that were mainly isolated from seeds or diseased legume plants. The experiments revealed that all Trichoderma strains isolated from soil of the Czech Republic showed mycoparasitic effect on se-lected pathogens. However, the efficacy of the strains depended on the type of fun-gal pathogen. Spore production of Trichoderma strains was also evaluated both in the control variant as well as the mutual interactions with pathogens. The results again showed differences in spore production among the Trichoderma strains. The myco-parasitic effect of the entomopathogenic fungi Isaria fumosorosea and Akanthomyces attenuatus against the pathogen Botrytis cinerea was also evaluated. The results re-vealed that although I. fumosorosea and A. attenuatus showed a significant mycopar-asitic effect against Botrytis cinerea, the mycoparasitic fungus T. virens showed the highest efficacy against the pathogen. The findings indicated that both mycoparasitic and entomopathogenic fungi are sustainable alternatives to artificial pesticides in the fight against fungal agents of plant diseases.
