During this thesis the localization of the prioritized proteins Tb927.11.13600, Tb927.11.14570, Tb927.4.840 and Tb927.6.4540 and their connection to the tripartite attachment complex of the protozoan parasite Trypanosoma brucei was examined to verify previous annotations found on the TrypTag database. Additionally, the essentiality of the prioritized proteins was evaluated. During this thesis the localization of the prioritized proteins Tb927.11.13600, Tb927.11.14570, Tb927.4.840 and Tb927.6.4540 and their connection to the tripartite attachment complex of the protozoan parasite Trypanosoma brucei was examined to verify previous annotations found on the TrypTag database. Modelling of tick borne enecephalitis virus (TBEV) proteins and their complexes

In this bachelor thesis the focus was on the study of non-structural proteins of the tick-borne encephalitis virus involved in the replicatory process of the virus. Non-structural protein number 3 (NS3) and number 5 (NS5) contain a helicase and an RNA-dependent RNA polymerase domain, respectively, and play a crucial role in the life cycle of the virus. Therefore, they are of interest to develop a potential antiviral drug inhibiting the replication of the pathogen. It has been found that these two proteins form a dimer during the replication of the viral RNA. The aim of the thesis was to identify the exact interaction of the two proteins with existing hydrogen-deuterium mass spectroscopy data as well as modelling the RNA into the complexes. The research was conducted by using computational biology tools including homology modelling using MODELLER, manual modelling in PyMOL and Chimera as well as Molecular Dynamics Simulations in GROMACS. Effect of different water regime on microorganisms of nitrogen cycle in permafrost

The gene abundances and community structures of nitrogen-fixing and denitrifying bacteria in permafrost-affected soils with different moisture level were analysed. Additionally, DNA- and RNA-based analysis was used to evaluate the variations in gene copy numbers and community composition between the total and active bacteria. The gene abundances of nitrogen-fixing and denitrifying bacteria were quantified by qPCR of the nifH and nosZ genes, and the gene copy numbers were compared between the soils with different moisture. Furthermore, the bacterial community structure variation was evaluated by analysis of the taxonomic classification obtained from sequencing data. Tato bakalářská práce se zabývá slovní zásobou amerického dialektu New York City English, která je výsledkem bohaté lingvistické krajiny této metropole. Vzhledem k tomu, že město New York má dlouhou historii imigrace, do řeči jeho obyvatel se dostalo mnoho exogenních slov, pocházejících z přistěhovaleckých jazyků. Kromě výrazů, které do města přinesli přistěhovalci, je newyorské lexikum bohaté také na slova a fráze, které vznikly přímo ve městě. Předchozí výzkumy provedené na toto téma ukázaly, že některé lexikální prostředky používané Newyorčany se liší od těch, které používají mluvčí ve zbytku Spojených států, a mnoho charakteristických rysů dialektu zůstává pro oblast New Yorku jedinečných. Dosud však bylo provedeno jen velmi málo nedávných výzkumů týkajících se rozšíření lexika mimo nářeční oblast a skutečného užívání této slovní zásoby mezi Newyorčany. Cílem této práce bylo tuto mezeru zaplnit. Rozšíření lexika mimo oblast města a jeho užívání bylo zkoumáno pomocí dotazníku, který byl zaměřen na tři skupiny respondentů: Newyorčany, Američany žijící mimo New York a obyvatele ostatních anglicky mluvících zemí. Údaje získané od jednotlivých skupin byly prezentovány, analyzovány a interpretovány. Výsledky průzkumu nejprve potvrdily, že Newyorčané ve srovnání s respondenty z jiných částí Spojených států a s respondenty z ostatních anglicky mluvících zemí obecně lépe znají slova, která mají kořeny v jazycích přistěhovalců. Také se ukázalo, že tuto slovní zásobu používají častěji ve svém každodenním životě. Pokud jde o výrazy, které vznikly přímo v New Yorku, výsledky ukázaly, že mnoho z těchto slov zůstává do jisté míry omezeno na oblast této metropole. Zjištění také naznačují, že některé ze zkoumaných slov typických pro tento dialekt se z řeči Newyorčanů postupně vytrácejí a zůstávají používané především mluvčími patřícími ke starším generacím. This bachelor´s thesis investigates the lexicon of New York City English (NYCE) dialect, which results from the rich linguistic landscape of the city. Due to the fact that New York City has a long history of immigration, many exogenous words have found their way into the speech of its residents. In addition to words that have been introduced into the dialect by immigrants, the New York lexicon is also rich for words and phrases that were coined directly in the city. Previous research conducted on this topic indicated that some lexical devices used by New Yorkers are different from those used by speakers in the rest of the United States, and many of the dialect´s characteristic traits remain unique to the area of New York City. To date there has been very little recent research done on the spread of the lexicon beyond the dialectal area and the actual use of this vocabulary among New Yorkers. This thesis has aimed to fill this gap. The spread and use of the lexicon was investigated by means of a questionnaire, targeted at three groups of respondents: New Yorkers, Americans living outside of New York City and people from other English-speaking countries. The data collected from each cohort were presented, analyzed, and interpreted. The survey results firstly confirmed that New Yorkers, in comparison with respondents from other parts of the United States and with respondents from other English-speaking countries, are generally more familiar with words that have roots in immigrant languages. New Yorkers are also more likely to use this NYCE lexis in their everyday lives. Secondly, as regards the expressions that were coined in New York City, the results revealed that many of these words remain to some extent restricted to the New York City area. Finally, the findings also indicate that some of the NYCE lexis surveyed is gradually disappearing from the speech of New Yorkers and remains known and used mainly by speakers belonging to the older generation.

Tato bakalářská práce se bude zabývat detailním porovnáním vybraných příběhů Sherlocka Holmese od Sira Arthura Conana Doyla se seriálovou adaptací televizní stanice BBC Sherlock vysílané během let 2010-2017. Seriálová adaptace je založena na knihách z 19. století, přesto se ale v mnoha podnětech liší a dějová linie se tak ve více částech naprosto odvrací od původních příběhů. Tím největším rozdílem mezi těmito dvěma adaptacemi je ale doba, do které je příběh zasazen. Zatímco původní příběhy se odehrávají v období druhé poloviny 19. století do počátku 20.století, seriálová adaptace je naopak zasazena do moderní doby 21. století. Mezi sebou budou porovnávány epizody Studie v růžové s románem Studie v šarlatové, Skandál v Belgrávii s povídkou Skandál v Čechách a jako poslední epizoda Šest Železných dam s povídkou Dobrodružství se šesti Napoleony. This bachelor's thesis offers a detailed comparison of three selected Sherlock Holmes stories written by Sir Arthur Conan Doyle with the BBC adaptation of Sherlock broadcast during the years 2010-2017. The series adaptation is inspired by the books from the 19th century, but they differ in many ways, as the storyline diverges completely from the original stories in many parts. However, the biggest difference between the two adaptations is the time period in which the story is set. The original stories take place in the period from the second half of the 19th century until the beginning of the 20th century, while the series adaptation is set in the modern times of the 21st century. The episode "A Study in Pink" is compared to A Study in Scarlet, "A Scandal in Belgravia" to the short story "A Scandal in Bohemia," and "The Six Thatchers" to the short story "The Adventure of Six Napoleons."

Předmětem této bakalářské práce jsou jazykové aspekty vyrůstání v bilingvním prostředí. Cílem práce je nahlédnout do života bilingvní rodiny, která zažívá raný bilingvismus u dětí, z pohledu rodičů i bilingvních jedinců a tyto dva pohledy porovnat. Součástí práce je teoretický úvod do problematiky bilingvismu u dětí a aspektů souvisejících s tímto tématem. Analytická část představuje provedený výzkum, v němž byly zkoumány zkušenosti s bilingvismem osmi rodin s dětmi, které se od raného dětství učí dvěma jazykům současně. This bachelor thesis focuses on the linguistic aspects of growing up in a bilingual environment. The aim of the thesis is to gain an insight into the life of bilingual families experiencing early bilingualism in their children, to reflect on these experiences from the perspective of both parents and bilingual individuals and compare these two views. The thesis includes a theoretical introduction to bilingualism in children. The analytical part presents the undertaken research, in which the experiences examining the bilingualism of eight families with children who learn two languages at the same time from early childhood were investigated.

A Comparison between John Steinbeck's Characters - George Milton from 'Of Mice and Men' and Mordeen Saul from 'Burning Bright'

Cílem této bakalářské práce je analýza postav z novel-divadelních her Johna Ernsta Steinbecka 'Of Mice and Men' (1937) a 'Burning Bright' (1950), s hlavním zaměřením na postavy George Miltona a Mordeen Saulové. Nejdříve se práce zaměří na život autora a díla samotná, což je, mimo jiné, důležité i jako základ pro analýzu postav. V rámci této kapitoly se zmíní primárně o motivacích Johna Steinbecka, probere vztah mezi již zmíněnými knihami a životem Johna Steinbecka, a poté se pokusí přiblížit odlišnost těchto děl. Dále následuje samotná analýza postav George Miltona a Mordeen Saulové dvěma způsoby: jejich vnější vzhled, a jejich psychologii a motivaci. Analýza nabízí různá fakta, jako například, že Mordeen a George sdílí jejich verzi Amerického snu s jejich společníky. Toto je dále rozvinuto pro prokázání podobnosti postav jako důkaz, že John Steinbeck používal opakující se charakterové rysy, témata a motivace ve své práci. The main aim of this thesis is the analysis of the characters from John Ernst Steinbeck's play-novelettes 'Of Mice and Men' (1937) and 'Burning Bright' (1950) with a focus on the characters of George Milton and Mordeen Saul. To provide a background for the analysis of the characters, the work first concentrates on the author and the works themselves. This chapter primarily centers on the motivations of John Steinbeck, provides a relation between the books and his life, and discusses the opposite nature of the two works. Subsequently, the work analyses the characters of George Milton and Mordeen Saul in two ways: their outer appearance, and their psychology and motivation. The analysis provides various facts, such as that Mordeen and George both share their version of the American dream with their companions. This is further developed to establish the similarities of the characters to prove that John Steinbeck had employed recurring character traits, themes, and motivations for his work.

Bakalářská práce se zabývá konceptem dvojího života, kontrétně v životě Oscara Wildea a v jeho díle Obraz Dorian Graye. Cílem práce je porozumět dualitě v autorově žitotě, ve vybraném díle a poukázat na jejich souvislost. První část představuje Wildeův život se zaměřením na jeho dualitu. Té si lze všimnout především v udržování dvou vztahů najednou. Jsou zde také popsány potíže spjaté s vedením dvojího života. Druhá část analyzuje typy dualit tří hlavních postav románu, s ústředním zaměřením na hlavního hrdinu Doriana Graye. Různé typy dualit jsou prezentovány prostřednictvím Basilovy homosexuality, veřejného vystupování Lorda Henryho, lišícího se od jeho tajných tuh, a soubojem těla a duše v případě Doriana, jehož dualita je navíc podpořena existencí portrétu. Práce se dále zabývá vývojem Doriana a osobami, které jej mohli ovlivnit včetně jeho dědečka Lorda Kelsa a Lorda Henryho Wottona. The bachelor thesis focuses on the concept of double life, concretely found in Oscar Wilde's life and one of his works - The Picture of Dorian Gray. The aim is to provide an understanding of duality in author's life and in the selected novel and show the connection between them. The first part of the thesis presents Wilde's life with the focus on the duality. His duality can be noticed mainly through leading two relationships at once. There are also shown the difficulties of leading a double life in this section. The second part analyses the types of dualities of the three main characters from the novel, with the main focus on the protagonist Dorian Gray. The different types of dualities are presented through Basil's homosexuality, Lord Henry's public image that differs from his secret desires, and lastly through the fight of the body and soul in the case of Dorian. The duality of Dorian is even supported by the existence of the portrait. Further, the thesis discusses the Dorian's development and how certain people could influence who he has become, including his grandfather Lord Kelso and Lord Henry Wotton.

Multispecies analysis of the role of epigenetic factors in aging

The following thesis provides the results of a multispecies analysis that was done to identify expression patterns of 209 genes encoding epigenetic factors that are associated with aging and longevity. The first part of the analysis investigated changes in the gene expression with aging for the long-lived species human and naked mole rat and the short-lived species mouse and rat. The second part aimed to further analyse the baseline expression levels of these genes in the long-lived species bowhead whale, human, blind mole rat and naked mole rat when compared to the short-lived species rat, mouse, marmoset and guinea pig. To achieve this, a number of Python scripts were developed, investigating the expression of epigenetic factors that potentially contribute to longevity.
