Paratrypanosoma is a novel early-branching trypanosomatid

2014 - anglický The kinetoplastids are a widespread and important group of single-celled eukaryotes, many of which are devastating parasites of animals, including humans. We have discovered a new insect trypanosomatid in the gut of Culex pipiens mosquitoes. Glyceraldehyde-3-phosphate dehydrogenase- and SSU rRNA-based phylogenetic analyses show this parasite to constitute a distinct branch between the free-living Bodo saltans and the obligatory parasitic clades represented by the genus Trypanosoma and other trypanosomatids. From draft genome sequence data, we identified 114 protein genes shared among the new flagellate, 15 trypanosomatid species, B. saltans, and the heterolobosean Naegleria gruberi, as well as 129 protein genes shared with the basal kinetoplastid Perkinsela sp. Individual protein phylogenies together with analyses of concatenated alignments show that the new species, here named Paratrypanosoma confusum n. gen., n. sp., branches with very high support at the base of the family Trypanosomatidae. P. confusum thus represents a long-sought-after missing link between the ancestral free-living bodonids and the derived parasitic trypanosomatids. Further analysis of the P. confusum genome should provide insight into the emergence of parasitism in the medically important trypanosomatids. 2016 - český Cílem předkládaného souboru publikovaných článků je charakterizovat sociální dovednosti učitele jako součást jeho profesní kompetence a vymezit jejich význam ve výuce angličtiny komunikační metodou v edukačních interakcích ve školní třídě. Teoretickými východisky výzkumů jejichž výsledky jednotlivé články předkládají jsou funkčně komunikační přístupu k jazyku a z něj vycházející komunikační metoda. Pozornost je věnována procesům interakce, komunikace a cizojazyčnému diskurzu ve školním prostředí s akcentem na cizojazyčnou výuku dospívajících. Na základě specifik cizojazyčné edukační interakce konkretizujeme sociální dovednosti učitele a vymezujeme sociálně-dovednostní aspekty cizojazyčné edukační interakce. Zjištění výzkumů prezentovaných v předkládaných článcích se týkají zejména žákovské reflexe edukačního stylu učitelů angličtiny a edukačního působení učitele v konkrétní třídní interakci. Výzkum je rovněž zacílen na analýzu vybraných sociálně-dovednostních aspektů edukační interakce a diskurzu ve výuce angličtiny komunikační metodou. Konkrétně se jedná o žákovské a učitelské iniciace komunikace, otázky a distribuci komunikačních příležitostí. The aim of the collection of published articles is to define the requisite social skills of the English language teacher as a part of his/her professional competence, in the context of communicative language teaching in classroom interactions between English teachers and adolescent pupils. The theoretical preliminaries of the research whose findings are presented in the articles are the functional communicative approach to language and communicative language teaching. The attention is paid to the processes of interaction, communication and classroom discourse at school with an emphasis on foreign language teaching. We try to specify social skills of the English teacher and define social-skill aspects of foreign language educational interactions. The findings of the research presented in the articles concern mainly the educational style of English teachers and English teacher's behaviour in particular classroom interactions. The research is also focused on selected social-skill aspects of classroom discourse within communicative language teaching, specifically, pupils' and teachers' initiations of communication, questions, and the distribution of communication opportunities. Predicting asp and pikeperch recruitment in a riverine redervoir.

2016 - anglický Recruitment of two species, asp (Leuciscus aspius) and pikeperch (Sander lucioperca), in a riverine reservoir was studied using a novel statistical approach. Both species are piscivorous and are stocked into reservoirs for biomanipulative purposes to reduce planktivore species. Long-term data series were used, but the number of potential predictors was high. Therefore, a novel informative statistical approach based on dimension reduction methods was applied. Quality of outputs was driven by sampling methods. Main factors affecting asp recruitment were zooplankton abundance, predator density and temperature. In terms of pikeperch fry measured with seine and trawls, the number of predators was the only important factor. Gillnets underestimate small fish and the data were unsuitable for statistical modelling. Faktory ovlivňující výskyt vodních makrofyt v nádrži Lipno

2013 - český This RNDr. thesis focuses on the species composition and cover in the littoral of the Lipno Reservoir in 2006. Aquatic macrophytes are an essential component of littoral communities in lakes. Their reasonable development positively affects biodiversity and stability of the aquatic ecosystems and water quality. The aim of this study was to characterise the development of littoral macrophytes in Lipno Reservoir, an artificial lake with seasonal water level fluctuation, and highlight controlling factors for macrophyte occurrence. The development of macrophytes was examined at 114 segments of the shoreline, each 50 m long, situated along the whole reservoir perimeter in regular 1km distances. The segments were surveyed for phytocenology, shore morphology, pedological characteristics of substrate, and anthropogenic impact. The survey identified 53 macrophyte species (24 hydrophilic terrestrial, 17 emergent,1 floating-leaved, 2 free-floating, 4 submersed, and 5 amphibious). The average vegetation cover of the examined segments was 14 %, with approximately one third of the segments without any macrophytes and another third with vegetation cover less than 10 %. Macrophytes occurred only in the uppermost eulittoral zone down to the level of 75 % probability of water surface exceedance. On the basis of a correlation analysis, the major factors affecting macrophyte occurrence in the littoral zone were shore morphology, light conditions in water and erosion effect of waves that together with water level fluctuation caused loss of fine particles from and degradation of the substrate in the littoral zone. Raman Microspectroscopy of Individual Algal Cells: Sensing Unsaturation of Storage Lipids in vivo

In particular, we have focused on lipid storage bodies and quantified the degree of unsaturation of algal lipids (iodine value). To verify our results, we developed a method for extraction of storage lipids from highly resistant Trachidiscus minutus cells and determined the fatty acid composition using GC/MS. We have found Raman microspectroscopy to be a fast, versatile, and virtually non-invasive tool for evaluating the potential of different algal species for algal lipid engineering and industry.
