Concepts of Space in George Eliot's Novels (Daniel Deronda)

Cílem práce je představit viktoriánské spisovatelky druhé poloviny 19. století z pohledu koncepce fiktivních románových prostorů. Práce nejprve stručně představí hlavní autorky viktoriánského románu ve společenském kontextu doby 19. století (sestry Brontëovy, George Eliot) a poté se bude věnovat analýze posledního románu G. Eliotové Daniel Deronda (ve srovnání s autorčiným dřívějším románem 'The Mill on the Floss') s důrazem na proměnu románového prostoru v díle, kterému dominuje židovská tématika, hledání a nalézání kořenů a tradic v osobním životě hrdiny. Práce se zaměří na evropský kontext románových prostor a tradic a zhodnotí význam posledního románu ve vývoji autorčiny tvorby. The aim of this diploma thesis is to introduce Victorian authoresses of the second half of the 19th century and concept of fictional novelistic spaces. Firstly, the thesis will shortly present the main authoresses of the Victorian novels in the social context of the 19th century (the Brontë sisters, George Eliot). Secondly, it will focus on the analysis of the last of George Eliot's novels, 'Daniel Deronda' (comparing it with her earlier novel 'The Mill on the Floss') with the emphasis on the changes of the concept of space in the novel, in which the Jewish theme dominates, and it will also describe searching for the roots and traditions in the personal life of the hero. Finally, the thesis will aim at European context of the concepts of novelistic spaces and it will evaluate the importance of the last novel written by George Eliot. Práce se zaměří na pojetí prostoru v současném britském románu; nejprve zmíní nejvýznamnější představitele současné románové tvorby v Anglii (McEwan, Graham Swift, Martin Amis) a stručně charakterizuje dobu, ve které romány vznikly. Poté se zaměří na průřez dílem Iana McEwana a na vybraných románech se bude snažit zachytit proměny McEwanovy tvorby směrem ke globalizaci prostoru jeho děl. Jádrem interpretační analýzy budou romány Betonová zahrada (The Cement Garden), Cizinci ve městě (The Comfort of Strangers), Amsterdam, Sobota (Saturday), Solar a Mlsoun (Sweet Tooth). Součástí kapitoly o prvním McEwanově románu bude srovnání s Goldingovým Pánem much (Lord of the Flies). My diploma thesis Space in Contemporary British Fiction will focus on the concept of space in contemporary British novel; at first, three of leading English authors will be mentioned (Ian McEwan, Graham Swift, Martin Amis), and it will briefly characterize the literary period in which their novels were published. In the chosen novels, the thesis will notice the changes of McEwan´s work towards the globalization of space. The core of interpretational analyses will be the novels The Cement Garden, The Comfort of Strangers, Amsterdam, Saturday, Solar and Sweet Tooth. The comparison with Golding´s Lord of the Flies will be a part of the chapter concerning McEwan´s first novel, The Cement Garden. Calcium signalling and gene regulation by carcinogenic Orai mutants

The main objective of this work is to define and characterize point mutations of the calcium channel Orai isolated form the large scale cancer genomic dataset. Those mutats were examined in the means of sub-cellular localization as well as functionality. Out of 17 investigated mutations, 5 were found to alter protein gating and switch the channel to partially or fully open state. Conformational changes of PsbQ protein from PSII studied by NMR

PsbQ is an extrinsic protein of photosystem II (PS II) playing a role in calcium and sodium homeostasis for oxygen evolving complex in photosynthesis. The solution structure of PsbQ in phosphate buffer including a short N-terminal helix has been resolved recently. The incorporation of PsbQ in PS II and its interactions with other PS II proteins were not elucidated yet. As a prerequisite for detailed interaction studies with other extrinsic PS II proteins such as PsbP, suitable conditions for nuclear magnetic resonance (NMR) investigations of both interacting partners should be established. In this thesis the stability of PsbQ in different buffers, as well as the influence of pH, temperature and salt concentration was investigated. Following stability tests, candidate buffer conditions were chosen for further NMR experiments exploring the influence on protein conformation and dynamics. Previously it had been found that Bis-Tris buffer provides suitable conditions for PsbP, while PsbQ had been investigated in phosphate buffer. In this work I established, that Bis- Tris buffer is appropriate for PsbQ as well, but that it may undergo structural and dynamical changes as evidenced by NMR experiments on recombinant stable isotope (N-15) labeled protein preparations. Some newly observed nuclear Overhauser effect (NOE) correlation indicate possible interactions from the N-terminal stretch to a loop region between the four stable helices of PsbQ. This indicates a less flexible solution structure than the one found in phosphate buffer, which still deviates significantly from the earlier crystal structure. Further studies are required to refine a three-dimensional structure based on these NOEs and some characteristic chemical shift changes. Subterranean rodents are interesting model group for studying of the circadian timing system due to their cryptic lifestyle and challenging living conditions. Tato práce mapuje osudy domorodých obyvatel Kanady v armádě v průběhu druhé světové války. Odpovídá na otázky, jaké motivy je vedly k účasti v této válce a jaký přístup k nim měla kanadská společnost po jejich návratu. Obsahem teoretické části je tak celkový vývoj vztahů mezi domorodými obyvateli a Evropany a druhá světová válka z kanadského pohledu s přihlédnutím k účasti domorodých obyvatel. Praktická část odpovídá na otázky zmíněné výše, rozebírá životy konkrétních domorodých vojáků a rovněž osvětluje přínos této práce pro české studenty. The main goal of this work is to trace down the fates of Aboriginal soldiers in the Canadian military during the Second World War and to answer the questions of what their motives were to enlist and how they were treated after coming home. Therefore, the theoretical part deals with the development of the relationship between the Aboriginals and the Europeans, summarizing the WW2 from the Canadian point of view while paying special attention to the presence of Aboriginals. The practical part answers questions raised above and stated in more detail in the Introduction. Further it deals with the biographies of concrete Aboriginals and the contribution of this thesis to Czech students. Until recently, it was assumed that African mole-rats search for food randomly, as it is in concordance with Aridity food distribution hypothesis. However, recent studies indicate that some subterranean rodent species including mole-rats could be able to use plant chemicals (kairomones) for food localization. In my master thesis I conducted a battery of experiments on four mole-rat species to prove, that these species also posses the ability of kairomone guided foraging independently of their social organization. Until recently, it was assumed that African mole-rats search for food randomly, as it is in concordance with Aridity food distribution hypothesis.
