Development of DiCre parental lineage of \kur{Babesia divergens }

2022 - anglický Babesia divergens is a tick-transmitted parasite of the order Piroplasmida, an important causative agent of bovine and human babesiosis. Even though Babesia represent an important worldwide veterinary threat and an emerging risk to humans, some of the fundamental principles of their parasitic lifestyle are still poorly understood. One of the approaches, which help us to better understand the biology of Babesia parasites and their interaction with hosts or vectors, is functional genomics. Hence, the aim of this thesis is to establish a DiCre parental lineage of B. divergens that may provide the absent genome editing technique of conditional gene knock-out. Such a tool will help to functionally characterize essential genes/proteins and validate them as novel targets for the yet missing specific treatment. We have selected and identified an eligible recipient locus in the B. divergens genome into which a DiCre cassette will be integrated via homology recombination. We have validated and determined the optimal concentration of WR99210 and G-418 for their use as selection markers for transfected B. divergens cultures. Finally, we have designed and constructed the plasmid vector containing the DiCre cassette for the generation of DiCre parental B. divergens lineage that may be universally applicable to functionally validate any genes/proteins of interest. PROSTITUTION IN THE CZECH REPUBLIC AND NORWAY A COMPARATIVE ANALYSIS

2013 - anglický Má diplomová práce se zabývá tématem prostituce. Jako strategii jsem vybrala kvalitativní výzkum. Pro vytvoření své práce jsem použila různé výzkumné metody jako například zúčastněné pozorování, nestrukturované rozhovory, případovou studii a kvalitativní srovnávací analýzu. Moje práce je založena na empirických výzkumech, které jsem uskutečnila v rámci stáže v organizaci Albertine Prosjektet ve Stavangeru a jako součást spolupráce s českou nevládní organizací Rozkoš bez rizika v Českých Budějovicích. Má zjištění vychází z nestrukturovaných rozhovorů se sociálními pracovníky z obou výše zmíněných organizací a s prostitutkami a následného propojení s teoretickými znalostmi. Mým cílem bylo vytvořit srovnávací studii prostituce v České republice a Norsku se zvýšeným důrazem na oblast Českých Budějovic a Stavangeru, abych mohla popsat současnou situaci na trhu s prostitucí. Feministické hledisko také hraje důležitou roli, protože prostituce může být vysvětlována jako genderový fenomén. Tato perspektiva je jedním ze stěžejních bodů mé práce. My thesis deals with the topic of prostitution. As a research strategy for my master thesis I have chosen qualitative research. For developing my paper I was using various research methods such as participant observation, unstructured interviews, case study and qualitative comparative analysis. My paper is based on empirical studies which I have obtained during my internship in the organization Albertine Prosjektet in Stavanger likewise on cooperation with the Czech NGO Rozkoš bez rizika in České Budějovice. My results appeared from unstructured interviews with social workers of both organizations as well as with women in prostitution, and subsequent interconnection with theoretical knowledge. My aim was to elaborate a comparative study of prostitution in the Czech Republic and Norway with predominant focus on the České Budějovice area and Stavanger area in order to present the current situation in prostitution markets. Feminist points of view play also an important role because prostitution can be explained as a gender-based phenomenon. This perspective is one of the key points of my paper. 2017 - anglický Diplomová práce je zaměřena na téma taneční kultury a dvorských festivit u dvora Alžběty I. Anglické. Intencí je souborně prezentovat informace o stupni naší informovanosti o tradici dvorských slavností u alžbětinského dvora, který proslul řadou zajímavých a stále jen do určité míry známých kulturních počinů. Cílem práce je zároveň porovnat kulturní situaci anglického dvora ve druhé polovině 16. století s ostatními dvory, popsat specifika a vlivy, které se v té době v Anglii vyskytovaly unikátně a které tam byly importovány. Práce se bude věnovat i fenoménu tzv. masques, které v Anglii tvořily specifickou formu dvorské zábavy s teatralizovanými tanečními výstupy. V neposlední řadě budeme uvádět i taneční a divadelní záliby samotné královny Alžběty I. a její postoj k tanečnímu umění jako důležitému prvku společenského vzdělání u dvora. The diploma thesis is focused on the theme of a dance culture and court festivities at the court of the queen Elisabeth I. The aim is to present information about the degree of our awareness about the noble festivities at the Elizabethan court that is very famous for number of the cultural achievements which are interesting but known only to a certain extent. The object of the thesis is to compare the cultural situation at the English court with the others in the second half of 16th century, describe specifics and effects which occurred in England originally and which were imported there in that era. The thesis will include the phenomenon named "the masques", which constituted a specific form of a court festivity with theatre-like dance performances. We will also cite the queen Elizabeth's fondness in dance and in theatre and her attitude to the dance art and court festivities as a significant element of the English nobility education in the renaissance era. Seizing the past in the Novels of Julian Barnes with Regard to the Author's Narrative Techniques

2022 - anglický Diplomová práce zkoumá pojetí minulosti ve vybraných dílech Juliana Barnese, přičemž tento jev uvádí do souvislosti s narativními technikami, které autor zvolil. Práce se zaměřuje na ústřední tematické okruhy: beletrizování historie a obousměrnou povahu minulosti a současnosti. Vytyčené okruhy odrážejí autorův přístup k uchopení tématu minulosti v jeho románech. Cílem práce je nejen rozebrat téma hledání historické pravdy, které je společné Barnesovům dílům, ale také vyobrazení minulosti ve vztahu k tématům paměti, pravdy a vlivu těchto faktorů na formování identity jedince i kolektivu. Zohledněna je nejen narativní, ale i tematická inspirace, kterou Barnesově současnému psaní poskytly postmoderní tendence, zejména pak skepticismus vůči historické pravdě a subjektivismus, s kterým na historii nahlíží. The thesis examines the concept of the past in selected works by Julian Barnes, regarding the narrative techniques chosen by the author. It focuses on central thematic strands: fictionalizing of history and the bidirectional nature of past and present. The outlined thematic strands represent the author's approach to treating the theme of the past in his novels. Besides the common theme of the search for historical truth that Barnes' works share, the aim of this thesis is to examine the depiction of the past in relation to the themes of memory, truth, and how these factors shape the formation of individual and collective identities. The thesis considers not only narrative but also thematic inspiration provided to Barnes' contemporary writing by postmodern tendencies, particularly the scepticism towards historical truth and the subjectivism with which he views history.
