Assessment of water mutagenicity

2013 - English Since severe health and environmental problems are caused by mutagenic organic pollutants there are urgent needs for the determination of the mutagenicity of surface waters. This work summarizes the literature regarding the occurrence and determination techniques of organic pollutants with mutagenic potential. Additionally, some selected organic pollutants were quantified in surface waters. For pre-concentration Blue Rayon, a sorbent based on a copper phthalocyanine dye suitable for the concentration of polycyclic aromatic compounds with mutagenic potential, was used. Vzhledem k některým vážným zdravotním a environmentálním problémům způsobeným mutagenními organickými polutanty je žádoucí stanovovat mutagenitu povrchových vod. Tato práce shrnuje literaturu zabývající se výskytem a technikami stanovení organických polutantů s mutagenním potenciálem. Navíc byly nalezeny vybrané organické polutanty v environmentálních vodách. Pro zakoncentrování bylo použito modré hedvábí, což je sorbent na bázi ftalocyaninu mědi vhodný pro zakoncentrování polycyklických aromatických sloučenin s mutagenním potenciálem. New potential antivirals against tick-borne encephalitis virus infection

2013 - English The experimental work of the thesis was concentrated on studying of the inhibition effect of twenty-six chemical substances on tick-borne encephalitis virus. The substances showing inhibition effect in High Throughput Screening assay were processed to Cytotoxicity assay, where their toxic effect on tissue cell cultures was determined. Subsequently, substances with inhibition effect and no cytotoxic effect were incubated with cell culture and virus in two different incubation time periods and the supernatant containing virions was examined using plaque assay. RNA biology of symbiotic bacteria in insects

2016 - English Non-coding RNAs (ncRNAs) represent an important part of bacterial genomes. However, only few studies RNAs with limited sampling were done concerning ncRNAs of insect endosymbiontic bacteria. This study provides a broad in silico genome sampling of insect endosymbionts (63 lineages of 27 genera) for ncRNAs and their modifications. Most strikingly it was found out that i) genes encoding modification enzymes conserved in particular genomes differ to high extent, ii) most of tRNA and rRNA modifications sites are conserved regardless whether the gene encoding the corresponding modification enzyme is conserved, iii) multiple endosymbiont lineages do not encode a full set of tRNAs. Our data imply that translation of endosymbionts is much less efficient compared to phylogenetically related free-living bacteria and that some of symbionts possibly need to cooperate with their cosymbionts or maybe even with their host to maintain translation. Nekódující RNA (ncRNA) představují d uležitou část bakteriálních genomů. Přesto se ncRNA endosymbiotických bakterií hmyzu dosud zabývalo pouze několik studií s omezeným počtem vzorků. Tato práce představuje in silico analýzu ncRNA a jejich modifikací pro široké spektrum linií endosymbiontů (63 linií patřících do 27 rod u). Za nejd uležitější přínosy práce považuji následující poznatky i) geny kódující modifikační enzymy konzervované v jednotlivých bakteriálních liniích se do zančné míry liší, ii) většina míst tRNA a rRNA modifikací je konzervována bez ohledu na to, jestli je příslušný gen pro modifikační enzym konzervován nebo ne, iii) některé linie endosymbiontů nekódují plný set tRNA. Naše data navrhují, že je translace endosymbiontů ve srovnání s jejich volně žijícími příbuznými mnohem méně efektivní a že někteří symbionti potřebují na translaci spolupracovat se svými kosymbionty nebo dokonce hostiteli. 2014 - English The bachelor thesis "The role of irrationality in selected Ian McEwan's early works" aims to identify and analyze subsequently the above issue in the early works of Ian McEwan. The thesis focuses on two short story collections, that is First Love, Last Rites (1975) and In Between the Sheets (1978), and on the 1978's novel The Cement Garden. In the theoretical part, postmodern literature is characterized in brief, with a focus on the question whether it is possible to label McEwan's work as "postmodern". The following practical part examines the theme of irrationality and analyzes the individual topics, using the author's selected works as references. Předmětem bakalářské práce "Úloha iracionality ve vybraných dílech Iana McEwana" je identifikace a následná analýza výše zmíněné problematiky v rané tvorbě Iana McEwana. Práce se zaměřuje na dvě povídkové knihy: První láska, poslední pomazání (1975) a Psychopolis a jiné povídky (1978), a na novelu z roku 1978, Betonovou zahradu. Krátce charakterizuje postmoderní literaturu a sleduje, do jaké míry je možné McEwanovu tvorbu zařadit do jejího kontextu. Hlavní, praktická část, zkoumá, jakým způsobem je téma iracionality ve vybraných McEwanových dílech rozvinuto a na příkladech konkrétních vybraných děl rozebírá jednotlivá témata. 2015 - English This bachelor work concentrates on the friendship of two significant writers F. Scott Fitzgerald and Ernest Hemingway who were part of an American group called the Lost Generation. The work is focused not only on their lives in light of their vicinity, but also on the exchanged correspondence of these authors. As such, it tries to find and, eventually, interpret the hidden purpose of their behavior and opinions. The conclusion summarizes the importance of the friendship of these literary giants with respect to their private and working lives. Bakalářská práce se zabývá přátelstvím dvou významných spisovatelů F. Scotta Fitzgeralda a Ernesta Hemingwaye, kteří patřili do americké skupiny zvané ztracená generace. Práce se zaměřuje nejen na jejich život z hlediska blízkého okolí, ale především na společnou korespondenci těchto autorů. Dále se v nich pokouší nalézt a posléze interpretovat skryté významy jejich chování a úsudků. Závěr práce shrnuje význam přátelství literárních velikánů z pohledu osobního i pracovního života. Elements of Transcendentalism in Chuck Palahniuk's Fight Club

2016 - English Transcendentalism, a literary and philosophical movement prominent in the 19th century in the United States of America, significantly influenced some of the subsequent literary movements. Some of the main tenets of Transcendentalism, like the belief in the power of an individual and rejection of authorities and materialism, are prominent in Chuck Palahniuk's novel Fight Club. The aim of this thesis is to investigate the portrayal of these social phenomena in selected Transcendentalist works and in Palahniuk's novel and to propose what could be some of the historical and social changes that influenced their depiction. Transcendentalismus, filozofické a literární hnutí 19. století působící ve Spojených státech amerických, znatelně ovlivnilo některé pozdější literární směry. Některé z hlavních principů tohoto hnutí, jako víra v individualitu lidí a nechuť k autoritám a materialismu, jsou zřetelné v románu Klub rváčů Chucka Palahniuka. Cílem této bakalářské práce je prozkoumat ztvárnění těchto společenských fenoménů ve vybraných transcendentálních dílech a v Palahniukově románu a předložit možné historické a společenské změny, které ovlivnily jejich zobrazení. 2016 - English Chaos and madness belong among the most prominent themes in the work of the American writer Edgar Allan Poe. For this thesis, I have decided to focus on these two themes and their relation to the settings which they take place in, mainly because the settings are a key feature in Poe's work. The connection of chaos and madness to the settings will be examined through four selected texts from the author's prosaic work. The aim of this thesis is to analyze these texts on the basis of these connections and therefore to provide new points of view on them. Šílenství a chaos patří k nejvýraznějším tématům v dílech amerického spisovatele Edgara Allana Poea. Ve své práci jsem se rozhodla zaměřit na tato dvě témata a jejich souvislost s prostředím, ve kterém se jeho povídky odehrávají, a to především proto, že prostředí hraje v Poeových dílech klíčovou roli. Vztah šílenství a chaosu k prostředí budu konkrétně zkoumat na čtyřech vybraných textech z autorovy prozaické tvorby. Cílem této práce je tedy analyzovat texty na základě těchto spojitostí a poskytnout tak nový pohled na věc. 2008 - English Emergency health service is a specific part of health care and in connection with ethical approach to non-cooperating patients many questions appear: To what extent is acceptable to force the patient to cooperate and to what extent is it necessary and important to leave the patient the right to decide on his or her health in life? Is it in accordance with ethics to tell the patient a white lie on his or her state so that he or she would be willing to cooperate? The aim my thesis is to find out which approach is ethically acceptable when treating patients who do not cooperate. Further I wanted to learn about the opinions of rescue workers on ethical dilemmas, which I described above. Záchranná služba je specifická součást zdravotnické péče a v souvislosti s etickým přístupem k nespolupracujícím pacientům se zde objevuje mnoho otázek: {\clqq}Do jaké míry je vhodné nutit pacienta ke spolupráci a do jaké míry je nutné a důležité ponechat pacientovi právo na rozhodnutí o jeho zdraví a životě?`` Je v souladu s etikou pacientovi říci tzv. milosrdnou lež o jeho stavu, aby byl ochoten spolupracovat? Cílem této práce je zjistit, jaký je eticky vhodný přístup při ošetřování pacientů, kteří nespolupracují. Dále zjistit názory zdravotnických záchranářů na etická dilemata, která jsem nastínil výše.
