Between Knowledge and Imagination (Travelogues in Literature of Czech National Revival)

2008 - Czech The study surveys the variety of travel literature in the period of Czech National Revival. It explores travelogues in contemporary book production, as well as in magazines and journals in the 1810s-1840s. Contemporary travel books appear to be opened to various discourses, for example science, philosophy, ideological and literary discourses. In consequence, travel literature could{\crq}ve develop in many forms with different functions or eventually the functions could blend together in a text. The study is focused on various movements and changes of travelogues, namely between fictional and non-fictional writing, and between popularity and art exclusiveness. Most of all, attention is dedicated to the imaginative possibilities of the genre. The specific imagination of the travelogues constituted in the tension of various poetics and styles and reflected their significant expressions and stylistic features. In this aspect, romanticism was of great importance to literary voyages. The analysis of original works, as well as of translated and foreign texts, has lead to questioning of the concept of national literature as single entity and in result to the idea that contemporary literature should be included in supranational context. Předkládaná práce mapuje různé podoby obrozenského cestopisu, které lze nalézt nejen v dobové knižní produkci, ale také v tisku v rozmezí desátých až čtyřicátých let 19. století. Na základě jednotlivých textů je zřejmé, že obrozenský cestopis byl značně otevřen různým diskursům: uplatňoval se v něm diskurs vědecký, filozofický, ideologický a samozřejmě i literární. Cestopisný žánr proto mohl nabývat zcela rozdílných funkcí, případně se v něm tyto funkce mohly vzájemně prolínat. Práce je intenzivně zaměřena právě k různým přechodům a proměnám cestopisu, a to zejména mezi literárností a neliterárností, ale také mezi populárností a uměleckou exkluzivitou. V centru naší pozornosti se přitom ocitly především imaginativní možnosti žánru. Specifická obraznost cestopisu se ustanovovala v napětí mezi různými dobově aktuálními uměleckými styly a poetikami a reflektovala jejich příznačné postupy a prostředky, přičemž zásadní význam měl pro literární cestopis především romantismus. Zohlednění nejen původních děl dobové cestopisné literatury, ale i překladových a cizojazyčných textů, jednoznačně náležících do obrozenského recepčního horizontu, zpochybnilo také úzce jazykově vymezený koncept národní literatury a poukázalo na nutnost začlenění obrozenské literatury do univerzálního (nadnárodního) kontextu. The Impact of the Destination Management to the Effectiveness of a Company

2015 - Czech The main aim of the dissertation was an identification of the role of a company on the horizontal and vertical level in the tourism management on the basement of analysis of subjects involved in the destination management. The aim of the work was also to propose a system model of tools and measures that will lead to an efficient participation of a destination management company on the management of a tourism destination and to increase of the efficiency of a company in tourism. The simulation model and the scenarios of the future development of the tourism destination, which are presented in the dissertation, create a value added above the main aim of the dissertation. The main methodological approach, which was used in the dissertation, was system dynamics. This method helped to reveal the complexity of a tourism system and to identify the connections among a number of stakeholders in the destination. The partial aim of the dissertation was also a comparison of the regions of South Bohemia and Upper Austria as tourism destinations. Not only the essential tourism indicators and figures, but also the systems of the destination management and a role of a destination management company were compared. The data from the above mentioned comparison were used in the proposed system dynamic model that simulates behaviour of the tourism destination represented by the Region of South Bohemia. Cílem disertační práce bylo na základě analýzy typologie subjektů, které působí v destinačním managementu cestovního ruchu, vymezit roli podniku na horizontální i vertikální úrovni řízení cestovního ruchu a navrhnout systémový model nástrojů a opatření, která povedou k efektivní participaci podniku destinačního managementu na řízení destinace cestovního ruchu a ke zvýšení efektivnosti podniku cestovního ruchu. Přidanou hodnotou nad rámec cíle práce je vytvoření simulačního modelu a následné simulace scénářů budoucího vývoje v destinaci cestovního ruchu. Základním vědeckým přístupem, který byl využit v disertační práci, byla systémová dynamika. Tato metoda pomohla odkrýt složitost systému cestovního ruchu a vazeb mezi množstvím stakeholderů v destinaci. Dílčím cílem práce však byla rovněž komparace regionů jižní Čechy a Horní Rakousko jako destinací cestovního ruchu. Srovnávány byly nejen základní ukazatele cestovního ruchu, ale také systém managementu destinace a postavení podniku destinačního managementu. Data z výše zmiňované komparace byla následně využita v systémově dynamickém modelu, který simuluje chování systému destinace jižní Čechy. 2015 - Czech The proposed thesis contributes to the basic knowledge in tick (Ixodes ricinus) and tick-borne pathogens (Borrelia burgdorferi sensu lato, tick-borne encephalitis virus) ecology in particular studying the spatial distribution, host associations and its causes and consequences in Central European habitats. Předkládaná dizertační práce se zabývá základním výzkumem v oblasti ekologie klíštěte (Ixodes ricinus) a jím přenášených patogenů (Borrelia burgdorferi sensu lato, virus klíšťové encefalitidy). Role of FtsH proteases in the cyanobacterium Synechocystis sp. PCC 6803

2015 - Czech This thesis focuses on the functional and structural characterization of FtsH proteases in Synechocystis PCC 6803. One of the aims was to determine localization and subunit organization of FtsH homologues in Synechocystis cells using GST and GFP tagged FtsH derivatives. The main result of the thesis is identification of two FtsH hetero-oligomeric complexes and one homo-oligomeric complex in Synechocystis cells. The large part of the thesis is aimed at establishing the role of the first hetero-oligomeric complex, FtsH2/FtsH3, in quality control of Photosystem II and at identification of a mechanism, how its substrate proteins D1 and D2 are recognized. Another part is dedicated to characterization of the second hetero-oligomeric complex, FtsH1/FtsH3, which consists of two essential FtsH homologues and which is here identified as an important regulatory element in maintaining iron homeostasis. 2016 - Czech This thesis is a compilation of six publications describing development of microsatellite markers for four mammal species and five insect species. Most of these species are endangered and the aim of developing microsatellites was their use in conservation-genetic studies. The thesis thus represents a contribution to methodological tools for investigation of endangered animals. Different approaches were used for the development of the loci, including inovative pyrosequencing, which represents a trend in DNA analyses with variety of practical aplications. Besides other things, this thesis documents increasing availability of the most modern molecular biology methods and their utilization in classical biological disciplines such as zoology, botany and ecology. Předkládaná dizertační práce je kompilací šesti publikací, popisujících mikrosatelitové markery čtyř druhů savců a pěti druhů hmyzu. Většinou se jedná o druhy ohrožené a cílem vývoje mikrosatelitů pro tyto druhy bylo jejich následné využití v ochranářsko-genetických studiích. Práce tak představuje obohacení dostupných metodických nástrojů k výzkumu ohrožených živočichů. K vývoji lokusů bylo využito různých postupů, včetně moderního pyrosekvenování, které představuje trend v analýzách DNA s celou řadou praktických aplikací. Tento text mimo jiné dokumentuje vzrůstající dostupnost nejmodernějších metod molekulární biologie a jejich využití v klasických biologických oborech jako je zoologie, botanika a ekologie.
