Mechanisms of dsRNA virus replication: Cloning, production and structural characterization of C-terminal domain of sigmaNS.

A gene fragment encoding the C-terminal domain of the sigmaNS protein of the avian reovirus was amplified via PCR. The amplicon was cloned using BsaI restriction enzyme into the pASKIBA37+ expression vector and transformed into One ShotTM TOP 10 Chemically Competent E.coli cells. Colony PCR was performed and plasmids were sent for sequencing. Sequence verified plasmids were transformed into One ShotTM BL21 (DE3) or Rosetta-gami B (DE3) Chemically Competent E.coli cells. During Pilot expressions the conditions for production of soluble recombinant protein were optimized and according to them the recombinant protein was produced in large scale. Stereotypes about bilingualism

The topic of this bachelor thesis is the phenomenon of bilingualism, specifically stereotypes about bilingualism and bilingual upbringing. The aim of the thesis is to describe these stereotypes and either disprove the existing prejudices or, on the other hand, confirm them as relevant. The thesis comprises a general introduction in bilingualism, predominantly in children, typology of bilingualism, theoretical knowledge about bilingual upbringing and, finally, strategies of upbringing in bilingual families. Also discussed will be the stereotypes and myths which are generally part of our notions about bilingual children. The empirical part of the thesis will analyze the speech of bilingual children up to the age of eight. Their speech will be used to test the verity of selected stereotypes and prejudices. Předmětem této bakalářské práce je fenomén bilingvismu, konkrétně určité stereotypy spojené s bilingvismem a bilingvní výchovou. Cílem práce je tyto stereotypy popsat, případné předsudky vyvrátit, nebo naopak potvrdit jejich pravdivost. Práce bude obsahovat obecné seznámení s bilingvismem, převážně u dětí, typologii bilingvismu, teoretické poznatky o bilingvní výchově a dále strategie výchovy v bilingvních rodinách. Budou též diskutovány stereotypy a mýty všeobecně zakotvené v našich představách o bilingvních dětech. Empirická část bude analyzovat mluvu bilingvních dětí do osmi let věku. Na jejich mluvě bude zkoumána pravdivost vybraných stereotypů a předsudků. A Comparison of Mario Puzo's The Godfather with the Film Adaptations

The student undertook an analysis of the varied aspects linked with The Godfather regarding plot, character presentation and dialogue, as well as aspects of the narrative less conducive to direct representation in the film productions. Then the student compared the three film adaptations with Puzo's text, comparing and contrasting the original novel with the three adaptations, referring to, among other aspects, how changes were adapted to issues pertaining to a cinematic audience and further developed by the renowned director and actors. The general consensus is that The Godfather I and The Godfather II, both winning academy awards, were vastly superior to The Godfather III. This comparison re-evaluates that perception. Student provedl analýzu rozmanitých aspektů spojených s románem Kmotr týkajících se děje, prezentovaných postav a dialogů, stejně jako aspektů příběhu méně vhodných pro přímou prezentaci ve filmové produkci. Dále student porovnaltři filmové adaptace s Puzovým textem, mimo jiné také způsob, jakým byly změnypřizpůsobeny problémům týkajících se filmového publika a dále rozvíjenyrenomovaným režisérem a herci. Dle mínění většiny jsou Kmotr I a Kmotr II, obaoceněné cenami akademie, výrazně lepší než Kmotr III. Bude zajímavé znovu totovnímání vyhodnotit na základě tohoto porovnání. This bachelor thesis focuses on the linguistic aspects of growing up in a bilingual environment. The aim of the thesis is to gain an insight into the life of bilingual families experiencing early bilingualism in their children, to reflect on these experiences from the perspective of both parents and bilingual individuals and compare these two views. The thesis includes a theoretical introduction to bilingualism in children. The analytical part presents the undertaken research, in which the experiences examining the bilingualism of eight families with children who learn two languages at the same time from early childhood were investigated. Předmětem této bakalářské práce jsou jazykové aspekty vyrůstání v bilingvním prostředí. Cílem práce je nahlédnout do života bilingvní rodiny, která zažívá raný bilingvismus u dětí, z pohledu rodičů i bilingvních jedinců a tyto dva pohledy porovnat. Součástí práce je teoretický úvod do problematiky bilingvismu u dětí a aspektů souvisejících s tímto tématem. Analytická část představuje provedený výzkum, v němž byly zkoumány zkušenosti s bilingvismem osmi rodin s dětmi, které se od raného dětství učí dvěma jazykům současně. Effect of different water regime on microorganisms of nitrogen cycle in permafrost

The gene abundances and community structures of nitrogen-fixing and denitrifying bacteria in permafrost-affected soils with different moisture level were analysed. Additionally, DNA- and RNA-based analysis was used to evaluate the variations in gene copy numbers and community composition between the total and active bacteria. The gene abundances of nitrogen-fixing and denitrifying bacteria were quantified by qPCR of the nifH and nosZ genes, and the gene copy numbers were compared between the soils with different moisture. Furthermore, the bacterial community structure variation was evaluated by analysis of the taxonomic classification obtained from sequencing data.
